以高效液相層析儀,分析防己中馬兜鈴酸Ⅰ、Ⅱ (aristolochic acid Ⅰ、Ⅱ)之含量。採用C18逆相層析管柱,移動相為乙腈-1.25%醋酸溶液(40:60),檢測波長為254 nm,得到良好之分析結果。馬兜鈴酸Ⅰ、Ⅱ在濃度0.63~40.00μg/mL、0.40~25.76μg/mL範圍內,利用上述分析技術進行對照標準品馬兜鈴酸Ⅰ、Ⅱ之分析得線性迴歸方程式及相關係數(r)分別為Y=1.06E-05X-0.28(r=0.9998),Y=8.28E-06X+0.12(r=0.9998),顯示良好線性關係。馬兜鈴酸Ⅰ之同日內及異日間之相對標準偏差為1.17%及1.71%,添加回收率為103.08±3.42%,馬兜鈴酸Ⅱ之同日內及異日間之相對標準偏差為1.28%及1.70%,添加回收率為103.13±3.68%。價購市售防己藥材二十件,利用上述高效液相層析法分析,、馬兜鈴酸Ⅰ含量介於0.030~0.192%之間,平均含量為0.086%,馬兜鈴酸Ⅱ含量介於0.017~0.118‰間,平均含量為0.043‰。
A high performance liquid chromatographic method for determination of aristolochic acid il and Ⅱ in Fangchi Radix was established. The samples were analyzed by HPLC on a Cosmosil 5C18-AR column and UV detector (254 nm) with acetonitrile-1.25% acetic acid (v/v) (40:60) as a mobile phase at a flow rate of. 1.0 mL/min. The regression equations of aristolochic acid Ⅰ and Ⅱ were Y = 1.06E-05X-0.28 (r=0.9998) and Y = 8.28E-06X+0.12 (r=0.9998), respectively. The intraday and interday relative standard deviations of aristolochic acid Ⅰ were 1.17% and 1.71%, and the recovery was 103.08±3.42%. The intraday and interday relative standard deviations of aristolochic acid Ⅱ were 1.28% and 1.70%, and the recovery was 103.13±3.68%. 20 samples of Fangchi Radix were analyzed. Results showed that average contents of aristolochic acid Ⅰ and Ⅱ in samples were 0.086% and 0.043‰, and the ranges.
