可拋棄式退化性關節炎電化學式尿液免疫感測器之研究

摘要


退化性關節(Osteoarthritis, OA)是現代人常見的慢性疾病之一,是一種對軟骨組織產生破壞分解反應的疾病,使骨頭與骨頭開始彼此磨損,是不可逆的病變,若能早期診斷對病情控制有很大幫助。採用OA相關生物標誌容易在病症初期被發現,本研究藉由量測尿液中OA相關的生物標誌濃度,來開發出非侵入式的OA電化學免疫感測器,可提升能長期監控、容易操作的可能性。 本研究以二茂鐵二甲酸作為電子傳遞媒子,利用自組裝單分子層方式將電子傳遞媒子固定在修飾電極上,利用循環伏安法與電化學阻抗法對電極表面進行逐層探討分析,並以化學電子光譜對修飾電極表面進行分析,分別在鐵氰化鉀溶液及三氯六胺釕溶液探討有無氧化還原媒子對電極電子傳遞速率之行為,結果顯示修飾二茂鐵二甲酸之電極有效提升電子傳的速率。 分別修飾上相對應抗體,將有無修飾電子傳的媒子之電極,用於退化性關節炎之三種尿液中之生物標誌 (COMP、TIINE及AAT)之感測,同時進行阻抗式及電流式的感測分析,建立電阻、電流對各種生物標誌之濃度的關係,最後並將此免疫感測電極應用在尿液檢體中COMP、TIINE及AAT之感測,成功地開發此可拋棄式非侵入式尿液退化性關節炎電化學免疫感測器。

Osteoarthritis(OA) is one of the common chronic disease. It is a cartilage tissue destruction decomposition reaction disease which cause wear between bones, and the lesions is unable to restore. Early diagnosis is very important. The use of OA associated biomarker can be found at the beginning of illness. Therefore, a long-term monitoring, non-invasive and easy-operated detection is essential. This study is to develop non-invasive immune sensors by measuring the concentration of OA related biomarkers in urine. In this study, 1,1-Ferrocenedicarboxylic acid, redox medium, was immobilized on the electrode surface utilizing self-assembled monolayer technique. The chemical structure was characterized by Electron Spectroscopy for Chemical Analysis. The electrochemical properties of the electrodes with and without redox mediator modification were characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry in ferricyanide and hexaammineruthenium chloride aqueous solution. The experimental results suggested the electron transfer rate was enhanced by the modification of redox mediator on the electrode. The electrodes with and without redox mediator were immobilized by antibodies and applied for the sensing of three OA biomarkers, COMP, TIINE and AAT, by impedance method and amprometric method. The correlation of impedance and current density to the concentration of each marker were established. It showed the high sensitivity and low detection limit. Those electrodes were used to detect the COMP, TIINE and AAT concentration in urine samples of healthy people and patients. The experimental results suggested that a highly sensitive disposable electrochemical immunosensor was developed successfully. For OA biomarker measurement, by using different biomarker in EIS analysis, it is found that the redox mediator modified electrodes has higher sensitivity and lower detection limits than non-redox mediator modified electrode. In the electrochemical impedance spectroscopy, we used redox mediator of modified electrode to sense OA biomarkers in urine. Either male or female, there are significant differences between the normal and OA patients in COMP and TIINE, OA patients have the higher concentration of biomarkers, but AAT as inflammation related marks, it is relatively non-specific. With the above-mentioned, this research successfully developed a disposable OA urine sensor.
