Inhibition of Nuclear Factor κB Associated with Hepato-Protective Effects of Chrysanthemum Morifolium in CCl4-Induced Liver Cirrhosis in Rats

抑制nuclear factor κB與菊花對四氯化碳於大白鼠誘導之纖維化之肝保護作用有關

摘要


研究動機:大多數之原發性肝癌與慢性肝炎以及肝硬化有關。乾菊花之頭部為東方之一種傳統藥物。在中國及日本,它被用作民俗草藥茶及用來治療眼疾。此草藥更有抗發炎、抗突變、以及多種抗微生物活性,如抗細菌、抗黴菌、抗病毒、以及抗螺旋體等活性。此研究之目的為探討菊花對四氯化碳於大白鼠誘導之纖維化之作用、並探討其作用機制。 材料方法:以四氯化碳於大白鼠誘導之纖維化探討菊花對肝之保護作用以及其作用機制。尤其對細胞損傷、細胞自戕、DNA損傷、纖維化nuclear factor κB (NF-κB)去活化之探討。 結果:菊花顯著地抑制纖維化、細胞自戕、降低血清之alanine aminotransferase (ALT)以及對NF-κB consensus sequence之結合。 結論:菊花有顯著之肝保護作用,而且抑制nuclear factor κB與菊花對四氯化碳於大白鼠誘導之纖維化之肝保護作用有關。就吾人所知,本研究為第一篇發現菊花有肝保護作用。

Introduction: Most of the primary hepatocellular carcinomas (HCC) reported worldwide are associated with chronic hepatitis or liver cirrhosis (LC). The dried flower-heads of Chrysanthemum morifolium (Compositae) are an oriental drug. They have been used as an herbal tea in Chinese folklore and have been used for the treatment of eye disease in China and Japan. In addition, this herbal medication has been found to possess anti-inflammatory, antimutagenic, and several anti-microbial activities, such as anti-bacterial, anti-fungal, anti-viral, and anti-spirochetal activities. The purpose of this study was to investigate the effect of Chrysanthemum morifolium (Cm) in CCl4-plus ethanolinduced cirrhosis and to determine the underlying mechanism. Materials & Methods: The hepato-protective effect of Cm on the animal model using CCl4-plus ethanol-induced cirrhosis was used. CCl4-plus ethanol-induced cirrhosis in rat was examined. In particular, the resulting cell damage, apoptosis, DNA damage, fibrosis, and nuclear factor-κB (NF-κB) inactivation were investigated. Results: Cm significantly suppressed fibrosis, inhibited apoptosis, and decreased the serum alanine aminotransferase (ALT) levels and binding to the NF-κB consensus sequence, which were markedly elevated in the CCl4-ethanol-treated group. Conclusion: Results suggested Cm possessed significant hepato-protective effect. Furthermore, inhibition of nuclear factor κB is associated with hepato-protective effects in CCl4-plus ethanol-induced liver fibrosis in rats. To our knowledge, this was the first study showing that Cm posses hepato-protective effect, in addition to its numerous well known traditional pharmacological effects.
