台灣雞源與鴨源分離之敗血性巴斯德桿菌之分子特性

Molecular Characteristics of Pasteurella multocida Isolated From Chickens and Ducks in Taiwan

摘要


本研究目的,是利用分子生物學方法進行台灣禽源敗血性巴斯德桿菌(Pasteurella multocida; P. multocida)之分子特性分析。自1995年1月至1999年6月,由本省各地發生家禽霍亂之病材,共分離57株P. multocida,包括鴨源41株、雞源16株進行試驗。以隨機增幅多型性DNA (random amplified polymorphi DNA; RAPD)技術,利用OPG 10及O PG 13二組引子進行分析,結果可將上述菌株共分成6 群。另按其所合質體之種類又可區分為6種類型,結合RAPD指紋圖譜和質體類型分析,共可細分為23種次分類。試驗結果顯示雞源與鴨源菌株間具有交叉感染之可能性,另外同一分子類型之菌株可在多個縣份同時發現,顯示其流行並無地域特性。此外,利用國外所發表可用以鑑定P. multocida 之PCR 診斷技術進行分析,發現此技術亦可適用於本省禽源之P. multocida,可將之與其它常見之革蘭氏陰性菌作區分。另以PCR方法證實本省禽類家禽霍亂分離之病原菌不具有敗血性巴斯德桿菌毒素(P. multocida toxin, PMT)基因,因此PMT之生成與禽類敗血性巴斯德桿菌之毒力並無直接關連。本研究結果可提供作為家禽霍亂分子流行病學和追蹤控制之參考。

This study employs molecular methods to characterize Pasteurella multocida (P. multocida ) isolated from chickens and ducks in Taiwan. Fifty-seven P. multocida isolates, including 41 duck isolates and 16 chickens isolates were collected and tested. All these isolates were isolated from January 1995 t o June 1999 in Taiwan. Random amplified polymorphic DNA (RAPD) technique using primers OPG10 and OPG 13 could divide these isolates into 6 distinct groups. In addition, these isolates could be divided into 6 groups based on the types of plasmids they contained. RAPD fingerprints together w it h plasmid profiles could further categorize these isolates into 23 types. Based on this category, we found that some types of isolates could in-feet both chickens and ducks. Moreover, the same types of isolates could be isolated in many different counties, indicating that the epidemic of these isolates were not limited locally. Polymerase chain reaction (PCR) reported previously for the identification of P. multocida was found to be applicable to Taiwan avian strains of P. multocida; this technique is useful for distinguishing P. multocida from other common G (一) bacteria. PCR also showed that P. multocida toxin (PMT) gene is absent in all isolates of P. multocida we tested, so PMT is not essential for the pathogenicity of these isolates. This investigation would be useful for the control and epidemiologic study of P. multocida in Taiwan.
