本研究利用固相萃取前處理及梯度移動相之高效液相層析方法,配合可見光檢測器,同時檢測食用油脂中四種蘇丹色素(Sudan Ⅰ~Ⅳ)。檢體經過固相萃取匣(alumina材質)淨化,再經RP18層析管柱(4.6×150 mm)與乙腈及0.1%甲酸混合溶液之梯度移動相分離,於波長500 nm偵測。添加0.1~5.0 ppm蘇丹一號、蘇丹二號、蘇丹三號及0.13~6.5 ppm蘇丹四號之4種蘇丹色素回收率分別為,蘇丹一號74.17~98.33%、蘇丹二號91.28~106.73%、蘇丹三號87.01~91.35%及蘇丹四號83.01~102.58%。食用油脂之4種蘇丹色素最低檢出限量均為0.05 ppm。本計畫共檢測包括辣椒油、沙拉醬及食用油脂等60件檢體,均未檢出4種蘇丹色素。
A HPLC method was developed for the simultaneous determination of four kinds of sudan dyes (Ⅰ, Ⅱ, Ⅲ and Ⅳ) in edible oils. Samples were first applied into an alumina SPE cartridge and eluted with 75% ethyl acetate in methanol. HPLC was performed as follows: a RP-18 column eluted with 0.1% formic acid/acetonitrile mixture as mobile phase in gradient at a flow rate of 1 mL/min. Sudan Ⅰ, Ⅱ, Ⅲ and Ⅳ in edible oils were identified and quantified with absorbance at 500 nm. None of these sudan dyes were detected in 60 edible oil samples.
