山櫻花B類功能開花基因之選殖與序列分析

Gene Cloning and Sequence Analysis of Type B Flowering Gene from Prunus campanulata

摘要


在被子植物花器發育的ABC模型中,B類開花基因包含PISTILLATA(PI)及APETALA3(AP3)兩個基因,主要調控花瓣與雄蕊的發育,與大多數開花基因相同,AP3和PI都屬於MADS-box家族基因。本實驗自資料庫中蒐集並且分析已發表確定其功能之植物B類開花基因序列,在其具高保守性的序列中設計引子對,以山櫻花(Prunus campanulata Maxim.)為實驗對象進行基因釣取,成功獲得山櫻花之AP3及PI全長基因,分別命名為PcAP3和PcPI。進行分析比對結果顯示PcAP3和PcPI皆具有典型的高保守性MADS-box,在親緣關係上此二基因與薔薇科植物最為相近。所得之山櫻花B類開花基因資料亦有進行蛋白質功能區塊和蛋白質結構之預測。本研究有助於進一步了解山櫻花各開花基因對花器發育之調控。

PISTILLATA (PI) and APETALA3 (Ap3) genes are the members of B class gene in ABC flowering development model. These genes regulate the normal development of flower petals and stamens. These two genes belong to the MADS-box family genes. In this research, we designed pairs of degenerated primers based on the conserved regions of AP3 and PI genes among different plants, then used it to get the full-length AP3 and PI genes from Prunus campanulata. After aligning with the other characterized B class genes, both genes we obtained contained a highly conserved MADS-box domain, and shared highly similarities with the same genes from other Rosaceae plants. Through this research, we found out that these two genes were very similar to those B class genes from other Rosaceae plants. Our research also provided valuable information on the prediction of the protein functional motifs and protein structure of these genes. It may help us to understand more about how the Prunus campanulata B class genes work in plant flowering.
