光線、培養基、酸鹹度及溫度對Lepiota naucina菌絲生長之影響

摘要


以馬鈴薯洋菜培養基(potato dextrose agar)培養Lepiota naucina菌絲時發現其在8℃以下或36℃以上時無法生長,而最適合生長之溫度爲24℃。在連續光照或光照與黑暗互相交替下較連續黑暗不利於菌絲生長,若以液體培養,測定酸鹹度由4.6至10.0時對菌絲生長之影響發現酸鹹度在5.3至9.2時均適合其生長,而以7.8爲最適合。菌種培養試驗時以麥粒爲材料最適合菌絲生長。在9種液體培養基中以下列培養基裝於250ml之三角瓶中,容量爲100ml時可得到最高之菌絲乾重量,培養基含硝酸鈣、磷酸二氫鉀、麥芽抽出物、硫酸鎂、氯化鉀、磷酸鐵、麥芽糖及蛋白腖。碳氮比爲30時最適合菌絲之生長。

Potato dextrose agar (PDA) was used to evaluate the effect of light and temperature on the mycelial growth of Lepiota naucina Fr.. No growth was observed lower than 8℃ or higher than 36℃ (optimal 24℃). Continuous darkness was found better than continuous light or 12-hrs alternating photoperiod. In a liquid medium, pH values ranged betweed 4.6 and 10.0 were tested and the mycelial growth was observed from 5.3 to 9.2 (optimal 7.8). The mycelial growth was found best on wheat grain among the 5 media tested for spawn. Among the nine liquid media, the highest mycelium dry weight was recorded in a 250 Erlenmeyer flask containing 100ml of medium C consisting calcium nitrate, monopotassium phosphate, malt extract, yeast extract, magnesium sulfate, ptassium chloride, ferric phosphate, maltose and peptone. The C/N ratio at 30 was found best for the mycelial growth of L. naucina.
