玉米莖腐病抗病品種之篩選方法

A Method for Screening Resistance to Pythium Stalk Rot in Corn Cultivars

摘要


本研究發展之玉米莖腐病(病原菌:Pythium aphanidermatum)抗病品種(系)之篩選方法,是將供試玉米種子播於六吋素燒陶盆中,每盆3株。於溫室中(24~36℃)長至40~45天,株高約1公尺時,進行接種試驗。接種時,以每克土約含30個卵孢子之壤土加水達於飽和含水量後,塗抹在供試玉米植株的莖基部(0~10cm),並以鋁箔包裹之,以防接種病土乾燥。將接種後之玉米置於氣溫25~35℃,相對濕度90%以上的溫室中3天,以促其發病。再經3天,即可記錄其罹病率。判定時,以該品種(系)玉米全部罹病倒伏者爲感病;部份罹病倒伏者爲中抗;全部未罹病者爲抗病。

An inoculation method for screening resistance to Pythium stalk rot (Pythium aphanidermatum) in corn cultivars was established in this study. Three seeds of each corn cultivar or line were sowed on a six-inch-pot and grown in a greenhouse at temperature between 24 and 36 C. After 40~45 days incubation until corn plants reached about 100 cm in height, they were used for inoculation. The artificial infested soil containing 30 oospores per gram of air-dry soil was saturated with water and smeared on the basal stalk (0~10 cm) of the tested plants. The inoculated site was wrapped with a piece of aluminum foil to keep the inoculum from drying out. The inoculated plants were incubated in the greenhouse with temperature at 25 to 35 C and relative humidity at 90% or more for three days. Thereafter, the environment was recovered to normal conditions for another three days. In general, cultivars (lines) susceptible to the fungus would readily lodge, whereas, those resistant ones stayed as healthy as non-inoculated plants. By this method, about 42% of 1100 tested corn cultivars (lines) were screened as resistant to Pythium stalk rot in our study.
