三島柴胡臺農1號之組織培養

Studies on Tissue Culture of Bupleurum falcatum L. cv. Tainung No. 1.

摘要


本研究之目的在探討三島柴胡臺農1號經由組織培養技術進行大量繁殖的可行性。試驗結果顯示,利用頂芽之緊殖效果較腋芽為優。以1/4量之Murashige and Skoog(1962)基本鹽類培養基配合1~2mg/1 Benzyladenine(BA)、0.1mg/1 α-Naphth-aleneaceticacid(NAA)、1% sucrose及0.9%之Difco-agar(pH 5.7),可促進由頂芽誘生之幼苗體在試管中生長,且有100%成活率。於培養基中另添加proline、tryptophan、glutamine及asparagine等氨基酸或椰子汁,不但能促進幼苗體之生長,且能減低幼苗之褐化率。液體培養基之頂芽緊殖率高於固體培養基,添加1mg/l BA之液體培養基在震盪條件下培養頂芽一個月,可有13倍之繁殖效果。

The purpose of this study was to develop an effective method for mass propagation of Bupleurum falcatum L. cv. Tainung No.1 through tissue culture techniques. The effectiveness of various explant parts and medium composition were investigated. Experimental results showed that the terminal bud (3mm in length) possessed a higher survival rate than lateral bud. For medium composition, 1/4 strength of MS (1962) inorganic salts with full strength of organic constituents, supplemented with 1 mg/1 BA, 0.1 mg/1 NAA, 1% sucrose and 0.9% Difco agar (pH 5.7) were ideal for the in vitro growth of shoot tips. Survival rate as high as 100% was recorded. Addition of amino acids, e. g., proline, tryptophan, and asparagine, or coconut milk to the culture medium could facilitate shoot growth as well as delay the occurrence of tissue browning. The propagation rate was higher in liquid culture than in solid medium. Thirteen adventitious buds could be produced from a single apex bud cultured in liquid medium and shaken at 100 rpm within one month.
