鰻(Anguilla japonica)腎臟與生殖腺組織之初級細胞培養

摘要


本實驗以鰻(Anguilla japonica)之腎臟、生殖腺(卵巢和精巢)做細胞的初級培養,結果指出較佳之培養液為Eagle's MEM和TC 199之等量混合液,加10%FCS。較佳之培養條件則NaCl濃度為0.137M,pH值為7.2~7.5,初植細胞密度為1.0×10^5 cells/ml~2.0×10^6 cells/ml,培養溫度為25℃等。 生殖腺在上述較佳的培養狀況下較腎臟容易長成monolayer,於培養後3~7天,卵巢與精巢皆可長成monolayer,長成monolayer之細胞皆為纖維樣、上皮樣細胞,用消化酵素0.1% trypsin-versene可繼代培養至第三代。

This report deals with the primary cultivation of kidney and gonadal tissues derived from the Japanese eel (Anguilla japonica). All fishes used were sexually differentiated. Cell cultures of the gonad appeared to be fibroblast-like and epithelium-like. It was found that the optimal temperature was 24℃, no growth of kidney cells occurred above 37℃ and the optimal pH of the medium (mixture of Eagle's MEM and Medium 199) ranged from 7.2 to 7.5.The proper initial cell density lay between 1.0×10^5 and 2.0×10^6 cells/ml. The ideal concentration of sodium chloride was 0.137M. Kidney cell cultures did not grow in higher salt concentration. Normal gonadal tissues had been successfully transferred three times.
