本研究探討溫度(15,25及35℃)、光照(1000及6000 lux)及培養液中NO3-N濃度(0.03及1ppm)對微細藻類Skeletonema costatum培養七天期間之脂肪酸組成與量之影響。結果顯示溫度及光照均顯著影響藻類脂肪酸組成。將藻類移至如上述設定之溫度、光照、NO3-N濃度後,其長碳鏈不飽和脂肪酸(PUFA)量,在1~2天內卽表現顯著差異。於35℃培養者其PUFA量比15℃及25℃者之量為低;培養達第五天以前,於高光照或25℃下培養者,其PUFA量均比低光照或15℃者為高。NO3-N濃度之影響,在本實驗中不顯著。不同溫度、光照及NO3-N濃度對S.costatum PUFA量之影響與細胞密度無關。
The present study investigated the combined effects of temperature(15,25 and35℃),illumination intensity(1000 and 6000 lux)and NO3-N concentrations of growthmedium(0.03 and 1ppm)on the fatty acid compsitions of marine phytoplankton,Skeletonema costatum.The alage were cultured for seven days after inoculation.Temperature and illumination intensity both significantly affected the polyunsaturatedfatty acid(PUFA)contents of the algae within the first and second culturing days,PUFA contents of the algae cultured at 35℃ were lower than that cultured at 15 or25℃.Before the fifth culturing day,algal PUFA was higher in the cultures of higherillumination(6000 lux)than in lower illumination(1000 lux).The same is also truefor temperature(25℃>15℃).The situation reverse after 5th day.NO3-N concentrationof the growth medium has no significant effect on the PUFA contents of thealgae.The results also indicated that the variations of algal PUFA content werenot related to cell density.
