The Application of the Taiwan Fleet Dynamics Management System to Fishing Fleets and Fishery Resources

本研究主要針對短期體外維持蝦血球細胞的培養條件進行改善,以利蝦子細胞性防禦機制的研究。來自淡水長腳大蝦(Macrobrachium rosenbergii)的血球樣本,分別培養在(1)MCHBSS培養基,於4℃下,30或60分鐘;(2)不同的培養基,於4℃下,30分鐘;及(3)L-15培養基,於4、25或30℃下,30分鐘。血球培養30分鐘後的活存率(53.3%)高於培養60分鐘(16.7%)者。使用不同培養基時,培養於L-15(63.5%)、L-15-胎牛血清(58.7%)、及L-15-蝦血清(55.7%)三種培養基的血球活存率無明顯差異;但以MCHBSS培養後,活存率明顯降低(36.8%)。當血球以L-15培養基於不同溫度下培養30分鐘後,培養在30℃的血球活存率(35.1%)明顯低於培養在4℃(56.1%)或25℃(52.6%)者。

This study intended to improve the short-term in vitro maintenance of hemocytes inorder to study cellular defense mechanisms in prawn.Hemocyte samples from Macro-branchium rosenbergii were cultured in(1)modified complete Hank's balanced salt solution(MCHBSS)at 4℃ for 30 or 60 min,(2)different media at 4℃ for 30 minutes,and(3)L-15medium at 4,25 or 30℃ for 30 min,respectively.A higher percentage of hemocyte viabilitywas obtained following hemocytes cultured for 30 min(53.3%)compared to culturedfor 60 min(16.7%).Using different media,there were no significantly difference of hemocyteviability between L-15 medium(63.5%),L-15-FCS(58.7%)and L-15-PS(55.7%);however,a significant decrease of viability was obtained following hemocytes cultured inMCHBSS(36.8%).Following hemocytes incubated in L-15 for 30 min at different temperatures,the viability was significantly lower at 30℃(35.1%)than at 4℃(56.1%)or 25℃(52.6%).
