Changes of Cortisol and Metallothionein upon Cadmium Exposure and Handling Stressed in Tilapia (Oreochromis mossambicus)

鎘處理與緊迫對吳郭魚(Oreochromis mossambicus)皮質醇和金屬硫蛋白之影響

摘要


成熟之雄性吳郭魚進行三種處理,處理組給予鎘注射、轉移緊迫;對照組給予注射生理石鹽水,與轉移緊迫;空白組則僅給予轉移之動作,經過0、6、12、24與48小時後,來比較各組實驗血漿之皮質醇與金屬硫蛋白之變化,探討這兩者緊迫反應因子彼此之關係。結果顯示:6小時注射處理後處理組與對照組之皮質醇含量顯著高於空白組,不論那一種處理,12小時後他們的皮質醇含量皆達最高,而在48小時後則處理組比另外兩組顯著高。金屬硫蛋白的含量則只有鎘注射組呈現時間效應,另外兩組直到第48小時才顯著比0小時增加,此增加之現象,我們推論可能是受到緊迫之誘導或是受到緊迫初期皮質醇增加之影響。

Tilapia (Oreochromis mossambicus) were divided into three groups: control (injected with saline and handling stressed), blank (handling stressed alone) and experimental group (injected with cadmium and handling stressed). Cortisol and metallothionein (MT) were determined in order to approach the inducing effect of MT by cortisol. Results showed cortisol level in experimental and control group was higher than blank group at 6 h after injection. Besides, it was significantly higher in experimental group than control and blank group at 48 h after Cd-injection. MT content was significantly higher during 6 h and 48 h than 0 h upon Cd-injection, but it wasn't showed on both of control and blank group except at 48 h. These data demonstrated the levels of cortisol were revealed at 6 h after injected-stress, the highest levels of cortisol were secreted at 12 h in this three groups and recovery after 48 h except experimental group. We suggested that MT content increase might be as an indicator of late stress response, but the major role of MT was greater appearing upon heavy metal treatment.
