靈芝菌種接種工具比較與太空包木屑培養基含量對靈芝子實體産量之影響

Comparison of Ganoderma Inoculation Tool and the Effect of Amount of PP Bag Sawdust Medium on the Ganoderma Fruiting Body Production

摘要


比較靈芝接種工具,即1.接種夾,2.接種匙,3.快速接種器接種24瓶(500ml/瓶)靈芝菌種所需時間;結果顯示依次需進360秒、120秒、及16秒,故特推薦此種作者自行設計之快速接種器供業者使用,些工具亦適用於香菇菌種之接種。 靈芝太空包以木屑培養基爲生長介質,其每包之裝填量影響靈芝子實體産量,故分三種處理,即 1.鬆軟包法(每包裝800g);2.半紮實包法(每包裝900g);3.紮實包法(每包裝1200g);對靈芝子實體産量之影響。共栽培76天,每處理各有48包,結果其産量鮮重依次爲224g、814g、及1,824g,乾重産量分別爲74.7g、271g、及608g,計算其物轉換率(BE%)分別爲0.65、2.09及3.52%,顯然偏低。建議應另選BE%高之品系,以提高靈芝産量。且生産靈芝子實體成功率依次爲14.6、45.8及100%,表示紮實包法不易被雜菌所污染,故推薦今後每靈芝太空包(長38.6公分,底面直徑10.6公分)應裝填木屑培養基1,200g,形成容積密度較高之菌包,可得較高子實體生産成功率而行較高産量。

Time needed for using 3 kinds of inoculation tools, i.e., forcep, spoon and ”Rapid Inocualtion Tool” to inoculate 24 bottles (500ml/bottle) of Ganoderma spawn was compared. Results showed that time needed were 360, 120, and 16 sec. respectively. Thus, ”Rapid Inoculation Tool” designed by the author was highly recommanded for the spawn or fruiting body production of Ganoderma This tool is also applicable to shiitake spawn inoculation. Amounts of sawdust medium in each PP bag affected the ”Cultivation Success Rate” (CSR%) of Ganoderma and its fruiting body production. Thus three treatments were compared as follows: 1. loose-packing method: each bag contained 800g of sawdust medium. 2. Semi-fully packing method: 900g sawdust medium was packed in each bag. 3. Fully-packing method: l,200g sawdust medium was filled in each PP bag. The Ganoderma in PP bag was cultivated for 76 days and 48 bags in each treatment. The results showed that the Ganoderma fruiting bodies were 224g. 814g, and 1,824g in fresh weight for 3 treatments. Dry weights were Rt.7g, 27lg, and 608g respectively. The biological efficiency (BE%) for them were 0.65, 2,09, and 3.52%. Comparatively, this BE% is low for Ganoderma tsugae Mum. There was a need for screening higher BE% Ganoderma strains for higher production of Ganoderma fruiting bodies. The cultivation success rates (CSR%) for three treatments were 14.6%, 45.8%, and 100%. This result indicated that fully-packing method could avoid the contamination of harmful fungi. Therefore, it is suggested that each PP bag (length: 38.6cm; diameter: 10.6cm) should fill in 1,200g of sawdust medium to form higher volume density of PP bag which would ensure higher CSR% and better production of Ganoderma fruiting bodies.
