細胞分裂素對火鶴花試管苗葉片培養再生之影響

Effect of Cytokinins on Plant Regeneration From in vitro Lamina of Anthurium

摘要


本試驗以火鶴花試管苗葉片爲材料,培養於添加不同細胞分裂素之1/2MS配方,結果以0.1、0.2 mg/L BA、或0.5 mg/L Kinetin處理之培植體產生的不定芽最多,約爲70%;而0.1 mg/L TDZ下,約有63%不定芽形成。供試之三種細胞分裂素亦會引起癒合組織產生;以添加0.1-0.5 mg/L TDZ癒合組織誘導率97.5-100%較高。在含有0.5 mg/L TDZ之培養基所產生之癒合組織最大。不同基因型不定芽的再生能力有差異。

Lamina of in vitro-grown Anthurium plantlets were used to study shoot regeneration. Half strength modified MS medium containing one of three cytokinins was applied. BA at 0.1-0.2 mg/L or Kinetin at 0.5 mg/L, caused 70% of the explants to regenerate adventitious shoots while 0.1 mg/L TDZ induced 63% explants to regenerate adventitious shoots. Callus formation was observed on basal medium supplemented with either cytokinin, especially with 0.1-0.5 mg/L TDZ, which induced 98-100% callus formation with larger calli. Regeneration abilities are statistically different among various genotypes.
