Cytokinins Stimulate Somatic Embryogenesis and Plant Regeneration from Male Inflorescence of Dieffenbachia 'Tiki'

This study aimed to explore the effect of cytokinins on increasing the somatic embryogenesis and subsequent plant regeneration of Dieffenbachia 'Tiki'. Male inflorescence explants isolated from spadix were cultured on a half-strength Murashige and Skoog (MS) medium, consisting of 2% sucrose, 1% glucose, 0.18% gelrite and 4 mg/L 2, 4 dichlorophenoxy acetic acid (2,4-D) for the basal medium, then add 0.5 or 1.0 mg/L N6-furfuryl adenine (Kinetin), 6-benzylaminopurin (BA) or Thidiazuron (TDZ). The percentage of somatic embryogenesis and number of mature embryoids increased with increasing concentration of cytokinins. The competency for inducing somatic embryogenesis is TDZ>BA>Kinetin in that order. Basal medium supplemented with 0.5 or 1.0 mg/L TDZ and 4 mg/L 2,4-D resulted in 100% somatic embryogenesis, and obtain an average of 35.2 somatic embryos per responding explant. This was the first report showing that some emblings developed from few mature embryoids in Dieffenbachia 'Tiki'.
