Genetic Diversity among the Populations of Calocedrus Macrolepis Kurz Var. Formosana (Florin) Florin

摘要


Calocedrus macrolepis Kurz var. formosana (Florin) Florin belonging to the family and genus of Cupressaceae and Calocedrus, respectively, is endemic to Taiwan,. We examined the genetic variation and relation of C. formosana, with the techniques of Inter-Simple Sequence Repeat (ISSR) fingerprints. One hundred and seventeen samples of C. formosana were collected from five natural areas. Eight ISSR primers were used to proceed the reaction test of PCR and 65 bands were obtained, of which 25 bands (37.3%) were polymorphic. The analysis of Molecular variance (AMOVA) revealed the variation was 31.7% (p<0.001) among populations and 68.3% (p<0.001) within population. Therefore, genetic differentiation within population and among populations were significant. Analysis of POPGENE showed a higher degree of genetic diversity value (H=0.3460) and genetic differentiation index (Gst=0.4307). But the gene flow among populations was (Nm=0.6610) indicating that the gene flow was restricted. The result of UPGMA cluster analysis showed that Deji, Hueisun, Mountains-Beiyu could be classified as a group; Bulon, Wulai fell into a group. The geographically categorized trend apparently was obvious. Geographical isolationwas an important factor to impede gene flow among different ethnic groups, resulting in greater genetic differentiation among them. This paper discussed the various reproductive and genetic differentiation among populations and gene flow under such situation in order to provide reference resource for genetic research of ethnic groups.
