臺灣豆馬勃屬新紀錄種與槲櫟實生苗之親合性探討

COMPATIBILITY OF A NEWLY RECORDED PISOLITHUS SPECIE WITH QUERCUS ALIENA SEEDLING

摘要


殼斗科植物多數具有外生菌根;本研究的子實體採自於嘉義縣中埔鄉殼斗科植物林下,此子實體所分離純化的菌株編號為Cms01;利用玻片培養可發現其菌絲形態具有扣子體;利用分子生物技術分析,其ITS序列經比對結果與Pisolithus orientalis (NR119745)相似度最高;並探討其與槲櫟(Quercus aliena)之親合性,經接種3個月後,證實此菌株能與槲櫟共生;利用電子顯微鏡觀察,可於其根菌結合體發現具有明顯菌毯及哈替氏網等外生菌根之特徵。綜合上述結果,證實此次分離的菌株應屬於台灣新紀錄種P. orientalis,並且能和槲櫟共生形成外生菌根,此亦證實此菌株為外生菌根菌。

The mycorrhizae of most Fagaceae species belong to ectomycorrhizae. In this study, the basidiocarps were collected from a Fagaceae spp. stand at Jhongpu area in Chiayi county. The no. Cms01 strain was isolated from the basidiocarp, it had found the clamp connection of mycelium by a slide culture, and the ITS serial alignments had high similarity with Pisolithus orientalis (NR119745) by molecular analysis. In aseptic synthesis experiment, this strain (Cms01) could form a mantle and a Hartig net in root associations with Quercus aliena by scanning electron microscope 3 months after inoculation. These results showed that this strain (Cms01) was a newly recorded species in the fungal flora of Taiwan, and it could form a typical ectomycorrhiza with Q. aliena seedling, and it also belonged to an ectomycorrhizal fungus.
