利用ISSR分子標誌探討刺花椒地區族群遺傳歧異度

Study on Genetic Diversity of Zanthoxylum simulans Local Population by Using ISSR Molecular Markers

Zanthoxylum simulans Hance is endangered member of Zanthoxylum L. (Rutaceae) on the central coast and in low-altitude areas in Taiwan. This study used the simple application of repetitive sequences (Inter-Simple Sequence Repeat, ISSR) molecular fingerprint technology. In this study, 108 individual samples were evaluated from the five areas of Sinfong, Houlong Dashan, Dongming, Tiejhenshan and Kinmen. We used the eight ISSR primers for PCR testing to gain 54 bands and 47 polymorphic bands. Analysis of POPGENE showed a higher genetic diversity value (H=0.359) and genetic differentiation index (Gst=0.248). Because the gene flow among populations was restricted, the result (Nm=1.523) was significant. The analysis of molecular variance (AMOVA) revealed the variation was 22.67% (p<0.001) among populations and 77.33% (p<0.001) within the population. Genetic differentiation within the population and among populations was significant (r=0.911, p=0.027(superscript *)). According to the geographically categorized trend, the result of UPGMA cluster analysis divided into two groups Taiwan and Kinmen. Regarding the result of the gene diversity and geographical relationship, we used the preserved strategy with in situ conservation and ex situ conservation to maintain the Zanthoxylum simulans local population.
