The Inhibitory Effect of Garlic on Cell Transformation Induced by DMBA

大蒜對DMBA所引發細胞轉形之抑制效能

摘要


雖然癌症的成因至今尚是個謎,但一般相信癌症是一個多發性來源所引起發的。正常細胞受到致癌物作用後,在初期即可能造成永久的改變,使細胞之分化程序受到干擾,進而引發細胞產生轉形作用,最後終至演變成惡性疾病。所以,細胞轉形作用可以說是正常細胞在進行轉變成癌細胞的必經過程。本實驗室,對於大蒜抑制倉鼠頰黏膜致癌作用研究,已頗有成效。本實驗之目的,即探討大蒜在活體外抑制細胞轉形的效能。首先,我們利用強致癌劑DMBA,建立其引發NIH3T3細胞產生轉形作用的模式做為控制組,然後分別將大蒜汁與DMEM培養基加上10%胎牛血清之溶液混合,並稀釋成0.5%、0.3%、0.1%及0.05%。受測細胞先經含有大蒜之培養基處理,然後再加入含0.5µg/ml DMBA之培養基,最後觀察並記錄細胞轉形點之數目。比較實驗組與控制組之結果後,發現濃度大於0.1%之含大蒜培養基能抑制DMBA所引發之細胞轉形作用,且此作用可以傳到下兩代之細胞。這表示大蒜之抗癌作用是在癌症發生的初期,且是在細胞DNA或胞核基質的階層上。對於將來進一步研究大蒜之抑癌機轉及發展大蒜成為癌症預防藥物,本研究提供了相當有價值之資料。

Although the etiology of cancer is still obscure, it is proposed that cancer develops from a multifactorial origin. Normal cells under the action of mutagens and carcinogens result in an initiation process whereby producing permanent changes in carcinogenesis and finally progressing to malignancy. Hence transformation is the process indicating that a normal cell is initiated and proceeds to become a cancerous cell. The anti-carcinogenesis effect of garlic has been demonstrated in our laboratory using the hamster cancer model. This experiment was thus designed to determine the inhibitory effect of garlic on cell transformation in vitro in order to explore the function of garlic on intracellular DNA. NIH3T3 cells were kept in culture, pretreated with garlic juice at different concentrations of 0.5%, 0.3%, 0.1%, and 0.05% in culture medium, and then stimulated with 0.5μg/ml 9, 10-dimethyl-1, 2-benz(a)anthracene (DMBA) to induce transformation. The efficiency of inhibition was evaluated by comparing the number of transformation foci in the DMBA control group with those in the garlic pretreated groups. The results showed that garlic at concentrations greater than 0.1% possessed significant transformation inhibition ability, and this ability of transformation inhibition could be inherent in the grand-daughter cells. It was also suggested that the effect of transformation inhibition appeared in the initiation stage of carcinogenesis, and the actions of garlic were exerted on the cellular DNA or at the nuclear matrix level. These information are highly valuable in further investigating the mechanisms of cancer inhibition of garlic and developing it as a cancer preventive agent in the future.
