臺灣產優雅星杯盤菌體外酵素之初步探討

Study on the Extracellular Enzyme of Asterocalyx Mirabilis in Taiwan

摘要


本研究利用固態培養基,進行核盤菌科的優雅星杯盤菌(Asterocalyx mirabilis)分泌體外酵素的特性分析,試驗使用Hankin與Anagnostakis兩氏之方法,測試該菌是否具有果膠反式消去酶(pectate transeliminase)、果膠去聚合酶(pectin depolymerase)和果膠酶(pectinase)。並進一步探討其澱粉分解活性(amylolytic activity)、脂肪分解活性(lipolytic activity)、蛋白水解活性(proteolytic activity)、磷酸酶活性(phosphatase activity)、尿素酶活性(urease activity)和幾丁質酶活性(chitinase activity);另以Yeoh氏等人之方法,測試該菌種的纖維質分解活性(cellulolytic activity);並以Pointing氏之方法測試木聚醣活性(xylanase)。於23°C恆溫培養7天,結果本菌種所產生的胞外酵素,包括:果膠去聚合酶、果膠酶、脂肪酶、纖維素酶、尿素酶、澱粉酶、蛋白質酶和木聚醣酶。

Solid media were used for the detection of enzyme production on different media from Asterocalyx mirabilis, Sclerotiniaceae. Hankin and Anagnostakis methods were used for the enzyme tests, including the enzymes of pectate transeliminase, pectin depolymerase, pectinase, amylolytic enzyme, lipolytic enzyme, proteolytic enzyme, phosphatase, urease, and chitinase. Cellulase were also detected by the method of Yeoh et al. Xylanase were further detected by the method of Pointing growing at 23°C for 7 days. There were pectin depolymerase, pectinase, lipolytic, cellulase, amylolytic enzyme, proteolytic enzyme and xylanase in the A. mirabilis but there were no pectate transeliminase, phosphatase, urease and chitinase.
