A Novel Suggestion on Clinical Determination of Metals in Whole Blood by Simultaneous Atomic Absorption Spectrometry

利用同時多元素原子吸收光譜法測定臨床全血中金屬的新建議

摘要


臨床上,經常利用測定血液中金屬的濃度,進行疾病、中毒和暴露於毒性金屬(例如鎘和鉛)的診斷,據所知,偵測臨床檢體中金屬的方法,仍然是以電熱式原子吸收光譜法最爲普遍,雖然電熱式原子吸收光譜法可提供較高的特異性及靈敏度,簡單的操作流程和最方便的樣品前處理,但每次的分析只能測定一種元素卻成爲最主要的限制。從電熱式原子吸收光譜法改良且已經成功發展出的同時多元素原子吸收光譜法,可提供偵測多元素的能力,亦可改善電熱式原子吸收光譜法主要的缺憾,進而降低分析時間和關於石墨配件所需的成本。在臨床實驗室測定全血中的金屬時,因爲全血樣品具有複雜的有機基質,所以測定金屬之前通常必須將全血消化,然而,爲了簡化分析流程和降低成本,期能以簡單稀釋的方式取代消化處理血液樣品,而利用同時多元素原子吸收光譜法測定金屬時,全血樣品確實可直接稀釋後上機進行測定,除此之外,同時多元素原子吸收光譜法更兼具同時測定多種元素的功能,以上兩種特性,使同時多元素原子吸收光譜法可以成爲臨床實驗室分析全血中金屬含量最具潛力的一種分析方法。

The diagnosis of diseases, intoxication and exposure to toxic metals, such as cadmium and lead, is frequently evaluated by determining their concentrations in whole blood. As is well known, electrothermal atomic absorption spectrometry (ETAAS) is still the most popular technique for the determination of metals in clinical samples. Although ETAAS provides the high selectivity, sensitivity, ease of operation and minimal sample preparation, the single-element capability is the major limitation to ETAAS. However, simultaneous atomic absorption spectrometry (SIMAAS) has been developed successfully to provide the necessary performance for multi-element determination and improve the main shortcomings of ETAAS, reducing analytical time and costs associated with the replacement of graphite parts. The whole blood samples were usually digested before the determination of metals due to the complex organic matrixes. Nevertheless, in order to simplify the analytical processes and reduce the costs, a simple dilution should be employed for the determination of metals in whole blood in clinical laboratory. Instead of digestion, the whole blood samples could be directly diluted for the determination of metals by SIMAAS. Providing two important characteristics of simple sample pretreatment and multi-element analysis, SIMAAS is the potential method for the determination of metals in the analysis of whole blood in clinical laboratory.
