Immunohistochemical Detection of Deleted - in - Colon - Cancer (DCC) Protein in human Normal Enterochromaffin Cells and Gastrointestinal Carcinoid Tumors

摘要


目的:DCC基因位於第18對染色體,在大腸直腸癌病患約有70%發生基因刪除。DCC基因攜帶一個屬於超科免疫球蛋白與N-CAM穿膜蛋白質相似,是一個推想中抑癌基因,本研究目的在以免疫組織化學染色法偵測腸胃道內分細胞及相關類癌腫瘤內DCC蛋白質。材料與方法:我們收集正常腸胃道以福馬林固定,石蠟包埋黏膜標本包括胃、小腸、盲腸、大腸及13例腸胃道類癌石蠟標本。本研究用兩種多株抗體用抗原恢復性免疫組織化學染色法偵測上述材料內DCC蛋白質及NSE之表現。結果:DCC蛋白質及NSE在正常腸胃道腸嗜鉻細胞及13例類癌均有表現。結論:我們認為腸胃道內嗜鉻細胞及類癌內含豐富DCC蛋白質。

Objective: A gene called deleted in colon cancer (DCC) has been identified on a region of chromosome 18. It is deleted in 70% colorectal cancers. The DCC gene encodes a protein belonging to the immunoglobulin superfamily with similarity to the N-CAM transmembrane proteins, and is a putative tumor-suppressor gene. This study was designed to verify the immunohistochemical expression of DCC protein in normal human endocrine cells of the gut and in related carcinoid tumors. Materials and Methods: We examined normal gastrointestinal mucosa from the stomach, small intestine, appendix and colon and 13 carcinoid tumors from the stomach, appendix and colon using DCC and NSE polyclonal antibodies with the antigen retrieval immunohistochemistry staining technique. Results: We detected DCC protein and NSE in enterochromaffin cells of normal gastrointestinal mucosa and all carcinoid tumors of the gastrointestinal tract. Conclusion: These results demonstrate that DCC protein may be abundant in entero-chromaffin cells and carcinoid tumors of the gastrointestinal tract.
