Molecular Phylogenetic Relationship Analyses of the Main Lineages of Nymphalinae (Rhopalocera, Nymphalidae) Based on Mitochondrial 16S rDNA Sequence

Fragments of the mitochondrial 16S rRNA gene of 8 species from Nymphalinae subfamily of the Nymphalidae were amplified and sequenced in this study. Combing with the homologous sequences of Nymphalidae from the GenBank, their sequence variations were analysed and their molecular phylogenetic trees were reconstructed with the neighbor joining, maximum parsimony and bayesian methods using the Argyreus hyperbius and Argyreus paphia of Argyrelinae as the outgroups. The results of sequence variations showed that the base compositions of their 16S rDNA fragment varied among these individuals as follows: A (41.8%), T (37.6%), C (7.8%) and G (12.8%), with a strong AT bias (79.4%) as usually found in other insect mitochondrial genomes. The phylogenetic trees showed that the Hypolimni is the sister group to the Melitaeini; the Melitaeini is a monophyletic group, in which the Eresia, Anthanassa, Tegosa and Phyciodes are closely related to each other. In addition, the results of phylogenetic analyses showed the tribe Nymphalini is not monophyletic, in which the Araschnia doris, Vanessa cardui and Symbrenthia lilaea constitute a monophyletic clade. However, the phylogenetic position of Junonia is not well-resolved in this study.
