Effect of the Compounds Purified From Chinese Herbal Drugs on Rabbit Platelet Aggregation (Ⅰ)

中草藥純化成份對兔子血小板凝集反應的作用(Ⅰ)

摘要


中草藥一直爲我中國傳統醫學在疾病治療上所取用的醫藥。而從中草藥純化出具生物活性的成份,亦是當前科學醫藥的研究重點之一。爲避免研究資源的浪費,極需建立有關中草藥成份之生物活性研究資料庫。本此目的,我們取得由中藥所純化的三十種純成份,並對它們進行生物活性的初步評估。我們是以兔子的血小板凝集反應作爲生物活性的分析模式。由實驗得到如下的結果:(1)山姜素、秦皮乙素、異土木香內酯、五味子乙素、東莨菪內酯和粉防己鹼,六種純成份在濃度爲100微克/毫升下,對膠原所誘發之兔子血小板凝集反應有大於90%的抑制作用。(2)六種對膠原刺激具抑制性的純成份,對花生四烯酸所誘發之血小板凝集反應也有中等程度以上的抑制作用;其中又以秦皮乙素和粉防己鹼的抑制性最強。(3)同樣的,六種對膠原刺激具抑制性的純成份,對腺瞟吟核苷二磷酸所誘發之血小板凝集反應也有中等程度以上的抑制作用;其中又以五味子乙素和粉防己鹼的抑制性最強。(4)對於血小板活化因子及凝血酵素所誘發的凝集反應,三十種純成份中只有粉防己鹼分別有85%及30%的抑制作用。前述對血小板凝集之抑制作用,都是濃度相關的。另外,(5)由實驗我們發現人蔘三醇本身就可以促使兔子血小板發生凝集反應,此反應具有一遲滯時間就有如膠原所誘發的反應。

The effect of thirty pure compounds isolated from Chinese herbal drugs on platelet functions were screened on washed rabbit platelets. Results indicated that 1) alpinetin, fraxetin B, iso-alantolactone, γ-schisandrin, scopoletin and tetrandrine inhibited the collagen-induced rabbit platelet aggregation to more than 90% at the concentration of 100μgml; 2) fraxetin B and tetrandrine (100μgml) also strongly inhibited the arachidonic acid-induced platelet aggregation: 3) 100μgml of γ-sc-hisandrin and tetrandrine could suppress the ADP-induced aggregation: 4) at 100μgml of concentration only tetrandrine inhibited platelet aggregation induced by PAF and thrombin to 85% and 30%, respectively: 5) in addition. panaxatriol (50-100μgml) induced platelet aggregation, which had a lag time similar to that of collagen.
