川芎嗪於大白鼠腦部各區之濃度分佈研究

Regional Brain Distribution of Tetramethylpyrazine in Rat

摘要


本研究應用一套靈敏、可靠的高效液相層析法,來定量川芎嗪於大白鼠腦組織各區濃度。以甲醇與水(50:50,v/v,pH3.0以正磷酸調整)當作移動相,使用碳18的道相分離管柱(4.6×250mm,粒經5μm),流速爲每分鐘1ml進行待測物分離,並以紫外光檢測器280nm爲偵測波長。腦組織最低可定量偵測極限爲0.1μg/g,每次取100μg來作分析。實驗以10mg/kg川芎嗪的劑量靜脈注射15分鐘後,迅速將大白鼠斷頭犧牲,並將其腦組織分成六個部分(划崗皮質、腦榦、紋狀體、海馬迴、,小腦及中腦),分析川芎嗪在各區之濃度,結果顯示,投藥御街組織六個部分的川芎嗪的濃度呈現均勻的分佈,且周邊血漿川芎嗪的濃度約高於腦組織五倍,靜脈注射後約17%的川芎嗪可以通過血腦障壁。

We used a rapid and reliable high-performance liquid chromatographic method for determination of tetramethylpyrazine in rat brain. The separation was achieved on a Cosmosil 5Cl8 column (4.6×250mm, particle size 5μm). The mobile phase consisted of methanol-water (50:50, v/v, pH 3.0 adjusted by orthophosphoric acid) at a flow rate of 1.0mL/min. Tetramethylpyrazine was monitored at a wavelength of 280nm. The lower limit of quantification in brain tissue was 0.1μg/g, and only a small amount of brain tissue (100 μg) was required for analysis. Concentrations of tetramethylpyrazine in various regions of the brain (cerebral cortex, brainstem, striatum, hippocampus, cerebellum and midbrain) were not significantly different at 15min after administration (10mg/kg, i v.). Mean concentration of the tetramethylpyrazine in plasma was approximately five times greater than in brain tissue. In additional analysis, about 17% of tetramethylpyrazine may penetrate blood-brain-barrier.
