Monitoring the Cellular Metabolism via Fluorescence Lifetime Imaging Microscopy

摘要


氧化磷酸化是細胞代謝裡一個重要的生化過程,且與生物體內能量生成與利用有關。參與過程的重要分子-NADH具有自發螢光的特性,使我們能利用螢光技術進行非侵入式的觀測及研究。在本研究裡,利用螢光生命期顯微技術觀察細胞代謝過程中NADH自發螢光生命期的變化,希望能建立生命期參數與細胞生理(細胞密度、凋亡與壞死)及病理狀態(阻斷電子傳遞鏈)間之關連性。

Oxidative phsphorylation is an important metabolic pathway that uses energy released by the oxidation of nutrients to produce adenosine triphosphate (ATP). Nicotinamide adenine dinucleotide reduced (NADH) participated in this process provides detectable fluorescence signal make it possible to utilize the non-invasive optical technique for further observation. In this study, fluorescence lifetime imaging microcopy (FLIM) is employed to monitor the function/conformational state of NADH associated with cellular metabolism in physiological (cell density, apoptosis, necrosis) and pathological states (inhibition of the electron transfer chain).
