利用離子配高效液相層析法同步分析綜合感冒藥劑中常見之八種主要成分

Simultaneous Analysis of the Eight Major Ingredients in Commerical Cold Medicine by Ion-Pairing High-Pressure Liquid Chromatography

摘要


Acetaminophen, aspirinum aluminum, chloropheniramine maleate, thiamine disulfide, caffeine anhydrous, codeine phosphate, potassium guaiacol sulfonate, ethoxybenzamide等為市售綜合感冒藥劑中較常見之主要成分,本文探討利用離子配高效液相層析法同步分析此八種成分之可能性,並探討配對離子濃度及pH值之影響。結果顯示使用Vercopak C-18(4.6x250 mm)層析管,並以含sodium 1-pentanesulfonate/sodium 1-heptanesulfonate (1:1;5mM,pH 3.0)之配對離子緩衝劑與甲醇作梯度沖提,流速為1毫升/分鐘,測定波長為280 nm可得最佳之分離效果。以此法分析一合成混合物(含以上八種成分)及市售三種感冒劑,可得到相當好的分析結果。

A method for simultaneous analysis of acetaminophen, aspirinum aluminum, chloropheniramine maleate, thiamine disulfide, caffeine anhydrous, codeine phosphate, potassium guaiacol sulfonate, ethoxybenzamid, the common ingredients present in cold medicine, was described. The method was based on reversed-phase ion-pairing high-pressure liquid chromatography. The best separation was achieved on a Vercopak C-18 column with a gradient elution using a solvent system containing an acetate buffer (1:1, sodium pentane sulfonate/sodium 1-heptanesulfonate, 5mM, with acetic acid to pH 3) and the methanol as mobile phase at a flow rate of 1ml/min, and a detection fixed wavelength of 280nm. The method gave excellent results for three commercial products and a synthetic mixture containing eight major ingredients. The effects of the counter-ion concentration and the pH on the separation were also discussed.
