黃條葉蚤(Phyllotreta striolata)傳播蕪菁輪斑病毒(Turnip ringspot virus)之初步探討

摘要


蕪菁輪斑病毒(Turnip ringspot virus, TuRSV)為新興的Comovirus 屬病毒。其與蘿蔔嵌紋病毒(Radish mosaicvirus, RaMV)特性極為相近,且均以十字花科作物為主要寄主。RaMV 於田間藉由黃條葉蚤(striped flea beetle,Phyllotreta striolata)傳播。本研究探討黃條葉蚤傳播TuRSV的可能性及其相關的傳播機制。研究結果顯示TuRSV可經由黃條葉蚤傳播,媒介昆蟲的獲毒取食時間(acquisition feeding time)短至10 sec即可獲毒,獲毒後的持毒時間 (retention period)可達14天,帶毒甲蟲傳播病毒的能力可維持6天。研究中以TuRSV 專一性引子對進行反轉錄聚合酶連鎖反應(RT-PCR)可專一且靈敏地檢測到單一蟲體上攜帶的TuRSV。

Turnip ringspot virus (TuRSV) is an emerging Comovirus that shares biological, morphological, serological and molecular properties with Radish mosaic virus (RaMV). RaMV is transmitted by striped flea beetle (Phyllotreta striolata) in the field. This study has shown that TuRSV, like RaMV, can be transmitted by P. striolata. The acquisition feeding time of the beetle vectors can be as short as 10 sec., and the viruliferous beetles can retain viruses for up to 14 days. However, the viruliferous beetles transmit viruses for only 6 days after acquired viruses. A specific primer set had been developed and can be used to detect TuRSV specifically and sensitively from a single viruliferous beetle.
