臺南地區魚塭綠藻之分子生物鑑定

Molecular Identification of Fish Farms Green Algae from Tainan City

摘要


本研究目的在於探討台南地區淡水與半鹽水至海水魚塭綠藻之分子生物鑑定。綠藻樣品採集自嘉南藥理科技大學鄰近魚塭、茄萣與四鯤鯓魚塭、七股及四草野生動物保護區等。共分離出10株淡水魚塭綠藻,及6株半鹽水至海水之魚塭綠藻,包括Chlamydomonas、Chlorella、Desmodesmus、Euglena、Monoraphidium、Mychonastes、Scenedesmus等7屬。經18S rDNA或ITS rDNA序列分析結果,以近鄰法(NJ)建構親緣演化樹狀圖,比對16株魚塭綠藻之ITS rDNA,皆發現ITS region rDNA可做為魚塭綠藻之分子標記。此外,亦發現ITS2 rDNA序列在綠藻的Desmodesmus與Scenedesmus兩屬的分類鑑定上,較ITS region rDNA(ITS1,5.8S,ITS2)及18S rDNA序列,更適合做為分子標記。

This study demonstrated the molecular identification of the freshwater and brackish to marine water fish farms green algae from Tainan City. Samples were collected from fish farm nearby Chia-Nan University, Jiading, Su-Kunshen fish farm, Chi-Ku and Szu-Tsao wildlife reservation in southern Taiwan. The phylogenetic trees were obtained by Neighbor-Joining method (NJ). Comparing the ITS region rDNA sequence between 16 fish farms green algae, 7 genera including Chlamydomonas , Chlorella, Desmodesmus, Euglena, Monoraphidium, Mychonastes, Scenedesmus, found that the ITS region rDNA can be used as DNA barcodes in molecular identification. Moreover, the ITS2 rDNA sequence demonstrated better than ITS region rDNA (ITS1, 5.8S, ITS2) and 18S rDNA sequences in identify both genera Desmodesmus and Scenedesmus.
