一個新穎的水稻"MADS-box"類似基因AK111796在種子發芽階段對缺水逆境之反應

Response of a Novel MADS-box-like Gene, "AK111796", to Drought-Stress During Seed Germination in Rice ("Oryza Sativa" L.)

摘要


實驗室初步從水稻根部生長與耐旱性的關聯性定位分析,發現一個MADS-box類似基因"AK111796"可能與耐旱性有關,為延續此發現,本試驗進行"AK111796"的特性分析及其對乾旱逆境的反應。序列分析結果顯示"AK111796"是一個新穎的MADS-box基因,可歸類在Mα基因次家族中,演化上最接近的基因為"OsMADS64"。為探討"AK111796"與耐旱性之關餘,試驗中以種子發芽時具有耐旱能力的台中私10號(TCSIO)與不耐旱的台南11號(TN11)為材料,分析"AK111796"以及其演化關條較接近的MADS-box轉錄因子對乾旱逆境之反應。當水稻種子在3% sorbitol處理5天與7天後,TCS1O不論根長或根數都比TNll多。基因表現分析結呆顯示,未處理或處理6% sorbitol時,TCSlO發芽7天後"OsMADS23"、"OsMADS25"和"AK111796"基因的表現明顯比發芽5天時提高,而TN11則無比現象。這些結果顯示水稻MADS-box基因可能和種子發芽時根的發育以及耐旱性相關,值得進一步研究。

Our earlier work on linkage mapping between root growth and drought-stress tolerance in rice revealed that the "AK111796" is a potential gene responsible for drought-stress tolerance. Sequence analysis indicated that "AK111796" is a novel MADS-box-like gene which could be classified into the Mα subfamily, and the OsMADS64 is the "phylogenetically closely related" gene in the MADS-box family. To further investigate the physiological and molecular responses of "AK111796" to drought-stress, two contrast rice cultivars were analyzed. Seeds of drought-tolerant cultivar Taichung Sen 10 (TCSI0) and drought-sensitive cultivar Tainan 11 (TN11) were germinated with or without sorbitol to evaluate their morphological and molecular changes in root growth and expression patterns of "AK111796". With 3% sorbitol treatment, TCS 10 has shown a better total number of roots and increased root length than TN11. In TCS 10, gene expression analysis showed that germination increased the expression of "OsMADS23", "OsMADS25" and "AK111796", no matter treating with or without sorbitol. In contrast, gene expression of MADS-box did not change in TN11. Taken together, these results provide new clues about the possible role of "AK111796" and "MADS-box" genes in drought-stress response and root development.
