市售常見香草植物抗氧化活性之評估

Evalution of antioxidant activity on commercial common vanilla plants

摘要


本研究目的乃使用市售常見的6種香草植物,以水或95%乙醇萃取,進行其萃取物抗氧化能力及活性之評估。分析項目包括:總酚化合物含量、超氧歧化酶(superoxide dismutase, SOD)活性、清除DPPH(1,1-diphenyl-2-picryl hydrazyl)自由基能力及總抗氧化能力。結果顯示,6種香草植物樣品萃取物之總酚化合物含量,其值介於19.79-255.97 mg GAE/g之間,含量前3高者,依序為馬鞭草乾燥品(乙醇萃)255.97 mg GAE/g、鼠尾草乾燥品(乙醇萃)169.83 mg GAE/g及甜菊乾燥品(乙醇萃)142.78 mg GAE/g。SOD活性方面,其活性值範圍為0-226.84 U/mg間,含量前3高者,依序為甜菊乾燥品(水萃)226.84 U/mg、甜菊乾燥品(乙醇萃)72.78 U/mg及鼠尾草乾燥品(乙醇萃)35.69 U/mg。清除DPPH自由基能力方面,雖然每種樣品清除DPPH自由基能力高低不一,但每種樣品皆呈劑量效應,隨著濃度的提高而增加,清除DPPH自由基能力較佳之前3高者,當濃度為50μg/mL時,依序為馬鞭草乾燥品(乙醇萃)37.70%、甜菊乾燥品(乙醇萃)19.90%及鼠尾草乾燥品(乙醇萃)18.41%。總抗氧化能力方面,每種樣品皆呈劑量效應,也隨濃度提高而增加,於低濃度下即顯示有良好清除ABTS.+自由基之能力,當濃度為50μg/mL時,清除ABTS.+自由基前3高者,依序為馬鞭草乾燥品(乙醇萃)27.24%、鼠尾草乾燥品(乙醇萃)8.56%及甜菊乾燥品(乙醇萃)8.36%。由以上結果顯示市售常見6種香草植物雖抗氧化成分及清除自由基的能力結果不一,各項香草植物萃取物樣品於低濃度下,即可發現亦有明顯的抗氧化效應,因此推測香草類植物仍具有良好的抗氧化能力。

There were 6 kinds of commercial common vanilla plants used in this study. The ways of extraction were used water or 95% ethanol, and evaluated the antioxidant capacity and activity of various samples. Analyzing items included total phenolic contents, superoxide dismutase activity (SOD), DPPH radical scavenging activity and total antioxidant capacity. The results indicated that the content of phenolic compounds in various sample extracts were between 19.79 - 255.97 mg GAE/g. The top three of contents of total phenolic compounds were 255.97 mg GAE/g of dried Verbena officinalis L.(ethanolic extracts), 169.83 mg GAE/g of dired Salvia officinalis Linn. (ethanolic extracts), and 142.78 mg GAE/g of dried Stevia rebaudiana Bertoni (ethanolic extracts), respectively. The SOD activity of vanilla plants were 0 - 226.84 U/mg. The top three of SOD activity in all samples were 226.84 U/mg of dried Stevia rebaudiana Bertoni (water extracts), 72.78 U/mg of dired Stevia rebaudiana Bertoni (ethanolic extracts), and 35.69 U/mg of dried Salvia officinalis Linn. (ethanolic extracts), respectively. In DPPH radical scavenging activity, despite the fact that the scavenging activity in each samples extract was different, it still found that all the samples had the dose response and enhanced with increase of concentration. The top three of commercial common vanilla plants, when the concentration was at 50 μg/mL, the DPPH radical scavenging activity of dried Verbena officinalis L.(ethanolic extracts) was 37.70%, dried Stevia rebaudiana Bertoni (ethanolic extracts) was 19.90%, and dried Salvia officinalis Linn. (ethanolic extracts) was 18.41%. In total antioxidant activity, all the samples showed the dose response and enhanced with increase of concentration. The top three of total antioxidant activity, which had good ABTS^(.+) free radical scavenging activity with a low concentration. When the concentration was at 50 μg/mL, the ABTS^(.+) free radical scavenging activity of dried Verbena officinalis L.(ethanolic extracts) was 27.24%, dried Salvia officinalis Linn. (ethanolic extracts) was 8.56% and dried Stevia rebaudiana Bertoni (ethanolic extracts) was 8.36%, respectively. From the above results show that the 6 kinds of commercial common vanilla plants which their antioxidant composition and the ability of scavenge free radicals were different, and that could be found a better antioxidant response with a low concentration. Thus, the study surmised that vanilla plants had the better antioxidant activity.
