冬瓜種子之TTC檢測法

摘要

以TTC染色結果作為判斷冬瓜種子活力的參考，檢驗程序設定如下：種子於20 ℃浸水24小時後，與種子間平行縱切，切除部份子葉，並剝除全部種皮及內膜。裸胚以0.5 % TTC溶液於30 ℃染色60小時，胚經TTC染色後，可分類為10種染色型式，與實際發芽率符合。本試驗仿照ISTA 種子檢查規則的格式，建議冬瓜種子TTC檢驗的研判準則為：染色後所染的紅色若為粉紅或種子胚間白色，胚與子葉連結部位染色不全或子葉斑駁等，皆判定為死亡種子。

Three varieties of wax gourd seeds were evaluated for the analysis of seed viability pattern. For this TTC (Tetrazolium chloride) testing procedure was followed. Seed were soaked in water for 24 hours followed by removal of testa and membrane inside. Then 1/3 of each soaked seed embryo was cut lengthwise. The cutted embryo were stained with 0.5% TTC solution at 30℃ for 60 hours. Nine patterns of TTC stained results were shown. According to ISTA (International Seed Testing Association) rule, the pink, white and mottled color seeds were considered as dead seed.
