蔗糖調節百合鱗片培養之再生能力與幼苗形態

Effects of Sucrose on Proliferation and Plantlet Morphology of Scale Culture in Lilium Speciosum Thunb. var. Gloriosoides Baker

摘要


本試驗測試培養基添加不同蔗糖濃度對豔紅鹿子百合溫室栽培鱗莖的鱗片、試管培養鱗莖的鱗片及試管培養鱗莖之再生能力與幼苗形態之影響。不添加蔗糖的處理培植體存活率、再生能力顯著降低,幼苗發育能力也減低。在溫室栽培鱗莖的鱗片或試管內培養鱗莖的鱗片方面,建議培養基蔗糖濃度皆為30 g/L,具有較高再生能力及幼苗可生成較多展開葉片。試管培養鱗莖試驗依以幼苗或鱗莖出瓶形態的不同,分別建議使用培養基蔗糖為60 g/L或120 g/L,60 g/L蔗糖進行培養生成的小苗具有較多的根數及葉片,較適合以幼苗形態出瓶培養,以蔗糖120 g/L進行培養可發育成較重的鱗莖,較適合以鱗莖形態進行冷藏處理後再栽植。

Plantlets producted from the scale explant derived from greenhouse and sterile culture of Lilium speciosum Thunb. var. gloriosoides Baker had different multiplication rate and morphology on MS basal media with variant concentration sucrose. None sucrose additive reduced the percentage of survival and regeneration ability of scale culture. Supplement of 30 g/L sucrose in medium for primary scale culture produced plantlet with 2 unfolded leaves, 2 roots and by 3.8 multiplication rate. On bulblet culture, we suggested that 60 g/L sucrose was a suitable concentration for seedling growth with vigor root, and 120 g/L sucrose could produce large bulblet.
