美鐵芋小葉培植體不定芽繁殖體系之建立

Establishment of Adventitious Shoot Regeneration System by Using Leaflet Explant of "Zamioculcas zamiifolia"

摘要


本研究利用美鐵芋小葉培植體進行芽體再生體系之建立,初代培養顯示小葉培植體經1%次氯酸鈉溶液表面消毒15min後,於MS(Murashige&Skoog)含80mg•L^(-1)腺嘌呤、1g•L^(-1)香蕉泥(基礎培養基)並添加2mg•L^(-1)TDZ之培養基中經八週初代培養,平均每個培植體可誘導6.3個不定芽。芽體大量增殖階段,以初代再生芽塊為次級培植體培養於基礎培養基添加1mg•L^(-1)BA組合1mg•L^(-1)TDZ之培養基中,八週後每個培植體可增殖17.5個芽體,平均芽長為12.9mm,可獲得質量皆佳的不定芽再生。小苗生根階段,將不定芽繼代於基礎培養基中添加0.5mg•L^(-1)NAA培養五週,發根率達83.3%,平均可獲致5.0條根;八週後發根率達100%,且根數增至15.3條。帶根之小植株出瓶後之存活率可達100%,經溫室栽培10週後有93.3%的小植株可抽出1.7個新梢,高度約80.2mm,具有5.1片完全展開小葉的獨立個體。本研究結果顯示,美鐵芋小葉段片是理想的培植體來源,經由芽體誘導、大量增殖、發根以及馴化條件的確立可提供芽體再生體系做為種苗大量繁殖之用。

An adventitious regeneration system of ZZ plant ("Zamioculcas zamiifolia" Engl.) was established by using leaflet section as explants in this study. The leaflets were sterilized with 1% sodium hypochlorite solution for 15 minutes before incubating on the Murashige and Skoog (MS) medium containing with 80 mg•L^(-1) adenine, 1 g•L^(-1) banana paste as basal medium and various concentrations of BA and TDZ in addition for primary culture. The results indicated that medium contain with 2 mg•L^(-1) TDZ had the best adventitious buds of 6.3 after 8 weeks of primary culture. A bud-clump section was used for multiplication stage, the explant was subcultured on the same basal medium supplemented with various concentrations of BA in combination with TDZ. 1 mg•L^(-1) BA in combination with 1 mg•L^(-1) TDZ, had the highest proliferation of 17.5 shoots per explants with 12.9 mm in height after 8 weeks of culture. During rooting stage, the in vitro shoot was cultured on the basal medium containing with 0.5 mg•L^(-1) NAA for five weeks of culture. A rooting percentage of 83.3% was obtained with five roots per plantlet. The rooting rate was up to 100% with 15.3 roots in average along with culture time up to 8 weeks. The in vitro plantlet with roots was acclimatized at 25℃ constant temperature in the growth chamber under greenhouse environment. After 10-week cultivation, results revealed the survival rate was up to 100% and 93.3% plantlet bore new shoots with 1.7 new shoots in average. New shoots were with 80.2 mm in height and with 5.1 fully expanded leaflets after 10-week culture in greenhouse. The present study revealed that leaflet segment is a proper explant source for "in vitro" adventitious shoot induction, shoot multiplication, rooting and acclimatization. The micropropagation system established in this study by using leaflet explant would provide ZZ plant nursery a useful seedling production system.
