在本研究中,主要為製備Gd(acac)TPP-Cremophor Micelles做為MRI造影藥劑。釓(Gd)為一順磁性金屬,能使周圍氫質子的縱向遲緩時間(T1)以及橫向遲緩時間(T2)縮短,以達到增強磁振造影影像訊號的目的。Gd(acac)TPP為一高脂溶性的化合物,假如不以介面活性劑-Cremophor EL將其修飾為水溶性,而直接將藥物由靜脈注射,會造成藥物阻塞在肺臟,無法達到造影的效果。研究中將Cremophor EL與Gd(acac)TPP 經過超音波震盪形成水溶性的膠粒(micelles)結構。實驗結果顯示所製備的Gd(acac )TPPCremophor Micelles具有高水溶性(log P為-0.93) ; R1值為16.964 (mMs-1)約為Gd (acac)TPP的二倍;粒徑大小約為10-18nm極適合做為活體造影之用。
Gd(acac)TPP-cremophor micelles were prepared as an MRI contrast agent. Gadolinium, a paramagnetic metal, can shorten the T1 and T2 values of surrounding protons, thus increasing signal intensity. Gd(acac)TPP is a highly relaxive, hydrophobic compound, and thus is unsuitable for in vivo administration. Gd(acac)TPP would accumulate in the lung if directly injected into the vein without modifying the surfactant-Cremophor EL. The Gd(acac)TPP was sonicated with Cremophor EL to form a hydrophilic “micelles” structure. The log P of Gd(acac)TPP-cremophor micelles is -0.93 with high water solubility; the longitudinal relaxivitives R1 is 16.964 (mMs)-1, or approximately twice the value of the corresponding parameter for Gd(acac)TPP; and the particle size is around 10-18 nm, which IS very useful for in vivo imaging.
