新開發的磁振造影對比劑Gd-TTDA-BOM與Gd-DTPA及Gd-BOPTA之比較:動物實驗研究

摘要


中文摘要 前言 本研究中主要在探討新開發之磁振對比劑Gd-TTDA-BOM與兩種商業化對比劑Gd-DTPA、Gd-BOPTA之比較。在3.0T的Magnetic Resonance Imaging(MRI)儀器中所展現的特性,以動物實驗來做影像分析,探討新合成磁振造影對比劑Gd-TTDA-BOM作為肝膽磁振造影對比劑的特性,以及與商業化的磁振造影對比劑做比較。 材料與方法 將本校醫藥暨應用化學系實驗室自行開發的Gd-TTDA-BOM合成後,與另外兩種已經商業化的磁振造影對比劑做比較。 選取9隻Sprague-Dewlwy(SD)大白鼠,由老鼠尾巴的靜脈注射將濃度0.1 mmol/kg body weight的Gd-DTPA、Gd-BOPTA、Gd-TTDA-BOM分別注射入三組正常SD 大白鼠體內,每組各有三隻大白鼠,觀察、分析這三種磁振造影對比劑在正常的大白鼠中的顯影特性,分別做動態掃描以及血管攝影的影像,比較Gd-DTPA、Gd-BOPTA、Gd-TTDA-BOM顯影效果的差異,測量三種顯影劑在正常的大白鼠肝臟、心臟、腎臟以及血管的影像表現,均以我們圈選的區域(ROI)加以測量,影像增強的程度以及各項數據,並持續觀察達注射顯影劑後一至兩個小時。 結果與討論 新開發的磁振對比劑Gd-TTDA-BOM在dynamic 以及angiography 兩種protocol,與商業化的顯影劑比較的結果,經過計算之後,我們可以看到,在心臟區域影像中,我們所得到Gd-TTDA-BOM的影像訊號強度不及兩種商業化的對比劑,但是在肝臟區域的影像,Gd-TTDA-BOM提供了很好的肝臟影像,經過影像數值的計算之後,相較於兩種商業化的對比劑,Gd-TTDA-BOM在肝膽系統擁有相當良好的影像增強程度(Enhancement)以及訊號雜訊比(SNR)。 結論 Gd-TTDA-BOM在肝膽系統中,的確是一個相當有開發潛力的磁振造影對比劑,未來還要建立起其腫瘤的動物模式,以及做進一步的毒性測試及生物體分布實驗,為其進入臨床試驗的路程鋪路。

英文摘要 Introduction: The purpose of this study was to compare the characteristics between a novel contrast agent Gd-TTDA-BOM and two commercial contrast agent Gd-DTPA and Gd-BOPTA in animal study using 3T MRI. Materials and Methods: MR images for normal and tumor rats were performed using 3.0T scanner. Dynamic MR images were obtained before and after intravenous injections of 0.1 mmol/kg Gd-DTPA or 0.1 mmol/kg Gd-BOPTA or 0.1 mmol/kg Gd-TTDA-BOM in normal rats. Sequential changes in signal intensity were measured with region of interest (ROI) from MR images , and the data aquired were analyzed to evaluate the enhancement and signal to noise ratio (SNR) in liver , heart , kidney , muscle and background. MR angiography was also performed after injection of similar doses of contrast agents. Results and discussions: The enhancement in liver of normal rats reached a plateau in 5-10 minutes after injection Gd-TTDA-BOM , maintained at least 2 hours, and gradually declined from kidney and liver. The enhancement in liver after injection Gd-TTDA-BOM was significantly higher than Gd-DTPA and similar to Gd-BOPTA. Conclusion: The preliminary results in this animal study indicated that Gd-TTDA-BOM has the potential of becoming a reliable liver MR contrast agent.
