不同運動量對大鼠阿基里斯肌腱修復的影響

Effects of Different Exercise Amount on the Recovery of Rats' Achilles Tendons

摘要


本研究目的是比較不同運動量對大鼠阿基里斯肌腱修復的影響。以54隻8週大體重200~250 g之Sprague-Dawley公鼠為實驗對象。隨機分控制組、走路15分鐘組(15分組)、走路30分鐘組(30分組),共三組;將大鼠右後腳阿基里斯肌腱切斷。控制組沒有任何運動介入,其餘兩組在傷後第5天進行走路運動介入,每天1次,運動時間分別為15、30分鐘;前2天速度4.0公尺/分鐘;之後12.0公尺/分鐘。分別在第5、12、19、28天評估阿基里斯肌腱功能指數(Achilles function index, AFI);傷後第28天犧牲大鼠,取出阿基里斯肌腱,觀察外觀、膠原蛋白生長情形、斷裂強度(最大破壞力量)(breaking strength, BS)、拉伸應力(tensile stress, TS)及恢復指數(recovery index, RI)。研究結果顯示:AFI值:第12天,30分組顯著優於控制組和15分組;第28天,30分組顯著優於控制組。直徑與膠原蛋白:各組受傷肢均顯著大於三組正常肢;在受傷肢中,30分組顯著小於控制組;有參與運動的組別膠原蛋白排列較整齊。斷裂強度:控制組受傷肢顯著大於正常肢;受傷肢間均未達顯著差異。拉伸應力:控制組受傷肢顯著小於正常肢;受傷肢間均未達顯著差異。恢復指數:三組間均未達顯著差異。本研究結論:本研究設定的模式下進行走路運動,可以促進新生膠原蛋白排列較整齊,提升修復後的品質;雖然增加運動量不影響斷裂強度及拉伸應力,但可能會使大鼠提早對受傷後走路的適應。

This study aimed to investigate the effects of different exercise amount on the recovery of rats injured Achilles tendon. Fifty-four Sprague-Dawley (SD) male rats (eight weeks old and 200 ~ 250 g) were randomly assigned to three groups: control, 15-minute (15 mins group) and 30-minute (30 mins group). The rats' Achilles tendon of right legs were cut first and then intervened by different exercise amount after the 5th day of Achilles injury and exercised once for each day. They exercised at the speed of 4 meters/min for first two days, and then the speed was increased to 12 meters/min. No exercise was performed in the control group. The functions of Achilles tendon were evaluated on the 5th, 12th, 19th and 28th day, and then all rats were sacrificed on the 28th day. The morphology, growth of collagen, breaking strength of tendon, tensile stress and recovery index were observed. Results: We found that AFI values on 12th day was significantly greater, 30 mins group was significantly greater than control and 15 mins. On the 28th day, 30 mins group was significantly higher than the control. Achilles tendon diameters in all experimental groups were significantly bigger than normal groups. In experimental groups, the control group was significantly bigger than the 30-min group. Collagens: Groups with exercise were found to be tidier. The breaking strength of tendon: Control experimental group was significantly higher than control normal. Experimental groups were not significantly different. Tensile stress: Control experimental group was significantly less than all the normal groups. Experimental groups were not significantly different. Recovery index: All the three experimental groups were not significantly different. In conclusion, the exercise program utilized in this study, we found that exercise amount won't affect breaking strength and tensile stress, but it may promote adjustability of walking and arrangement of newly collagen more order after injury.
