磁性粒子於醫療及酵素固定化、純化之應用

Magnetic Particles Applied on Medical Field and Immobolization, Purification of the Enzyme

摘要


近年來,磁性載體技術備受重視,是以未磁化粒子做為基質,給予磁場後轉變成具有磁性的載體,其表面藉由官能基適當修飾後,進一步與無磁性目標物質結合,於外加磁場下達到分離效果。磁性奈米粒子的製備與加工製程的進步,但使其應用價值大為提高,如應用於醫療診斷,DNA、RNA純化,分析領域,磁性藥物傳遞,廢水處理及蛋白質、酵素固定化等。本文中將介紹近年來一個新興的研究主題:磁性粒子於醫療及酵素固定化、純化之應用。文中將舉這幾年來以磁性粒子應用於酵素純化過程中之作用機制、以及應用於多種酵素純化之研究成果為例,說明磁性粒子作酵素純化載體之可行性。冀望能讓讀者瞭解此新興議題之發展及未來對於酵素純化技術之助益性。

In recently years, the development of magnetic carrier technology was to use the un-magnetic particles as matrix, and provide a magnetic field to transform into the magnetic support, after modifying the surface of magnetic carrier by suitable functional, the enzyme was immobilized onto a support. The magnetic carrier technology is already used in medical diagnostic, such as the contrast agent could be made from magnetic nanoparticles, and it was treated with magnetic resonance imaging (MRI), the diagnostic of cancer occurrence could be observed from the symptom of the malignant cel1. In the app1ication of the magnetic drug targeting, the magnetic microballoon was used as the carrier, and it could be injected to the animal, the microballoon will aggregated in the area of the malignant cell by the external magnetic field, it could increase the efficiency of the medical and decrease the side-effect of the kidney. In the hyperthermia, the temperature increment will be faster by the conduction of the magnetic particles, and it will kill the tumor cell easily. In the others, the coating of photosensitier on the magnetic particles could be applied in the photodynamic therapy (PDT), and the magnetic carrier technology is also wildly used in the purification of DNA, RNA and enzyme purification.
