豬腦萃取物對保護神經細胞抗類澱粉蛋白毒性之研究

Protective Effects of Porcine Brain Extracts Against Neuronal Damage by Amyloid Peptide

摘要


本研究乃以豬隻屠宰場之副產物-豬腦為原料,分別使用酵素水解與溶劑萃取濃縮製成豬腦萃取物(Porcine Brain Extracts, PBE),經由PC12神經纖維母細胞評估PBE抑制Aβ40毒性之試驗,包含細胞存活率(MTT assay)、發炎反應(ROS, NO)及氧化壓力(超氧陰離子, Catalase)。結果顯示,以0.1~10μM等不同濃度之聚合Aβ40處理PC12細胞後可發現細胞存活率隨著Aβ40濃度的提升而降低,然不同濃度(10、50及100μg)之PBE皆能提升細胞的存活率達50.3%至85.1%,與單獨經Aβ40處理的組別(42.5%)比較有顯著差異。此外,PBE可顯著降低因Aβ40促進發炎而上升之ROS與NO之生成量,改善發炎現象。另於細胞內之氧化壓力變化,PC12細胞經Aβ40處理會提高其超氧陰離子的生成達100%,並降低Catalase 75%之活性;但隨著PBE濃度的增加則可顯著降低Aβ40對神經細胞所引起之氧化壓力。綜觀上述,PBE可藉由其具抗氧化與抗發炎之作用,保護神經細胞抵抗類澱粉蛋白所誘發之神經毒性。此研究發現可進一步作為動物試驗之基礎,具有開發作為減緩神經退化性疾病的健康食品之潛力。

The purposes of this study were to investigate the functional characteristics of porcine brain extracts (PBE) which from slaughter house byproducts. We extracted PBE by enzyme hydrolysate and solvent concentrate, and then examine the effect of Aβ40 induced cytotoxin for cell, including cell viability, inflammation expression and oxidative stress were carried out in this study. We founded that cell viability on PC12 cell decreased by increasing Aβ40 concentration (0.1-10 μM), while increased to 50.3~85.1% by various concentration (10, 50 and 100 μg). According to the results of oxidative stress in PC12 cells, the content of super oxide anion increasing to 100% degree and reduce Catalase activity for 75% after Aβ40 treatment; which significantly decrease under the increasing concentration for PBE. These results indicated that PBE can against neuronal damage for Aβ40 induced cytotoxin by anti-oxidative activity and anti-inflammation. These results can be further research on animal model experiments for functional food development of neurodegenerative diseases.
