薑黃素自乳化液配方、載量及釋放特性評估

The Evaluation of Self-emulsifying Delivery System(SEDS) on Curcumin Loading and Its Vitro Simulated Releaseing Contents

摘要


薑黃素為近年來十分熱門之保健、營養素材,有許多研究文獻證實其保健功效,然而薑黃素常因低水溶性、低生物利用率及低穩定性,在利用上受到限制,如何提高溶解性成為研究主題;〝自乳化"為一種脂質載體系統,能增加脂溶性成分之溶解度、生物利用率及穩定性。本篇研究擬利用不同油脂、乳化劑、助溶劑,透過自發性、均一性、分散性篩選出最高油脂含量自乳化配方,探討不同薑黃素載量對自乳化液的品質影響及模擬胃腸中釋放情形之評估,其實驗結果如下:乳化劑以脂肪酸山梨醇酐酯(Span 80)與藻油、葵花油、橄欖油、棕櫚油等均具有較好的互溶性,配合聚山梨醇酯八十(Tween 80)能有最佳自發性、分散性、均一性,並以Tween 80與Span 80以1 : 1之比例與實驗所使用之油脂混和能有最高載油量,並和棕櫚油混和能達最高載油量66%及最小粒徑267.7 nm;薑黃素在試驗配方中溶解度為6.37 mg/mL,在自乳化液配方中添加3、4.5、6 mg/mL之薑黃素含量,其粒徑、濁度大小隨薑黃素含量之提升而有上升之趨勢(p < 0.05),並於添加6 mg/mL有最大粒徑334.0 nm,在模擬胃腸釋放特性試驗中可發現在胃液、腸液中分別釋放量為69與80%,未來如何減少胃中釋放為另一研究重點。

Curcumin, a nutrition material, is known for its health ingredients in recent years. There are many research have already confirmed that it content a various benefits for human health. However, the application of curcumin is still limited due to its low water solubility, low bioavailability and low stability. Therefore, how to improve its solubility, bioavailability and stability is an critical research direction to the related scientist. The subject of the study; self-emulsifying delivery system (SEDS) is a lipid-based carrier that increases the solubility, bioavailability and stability of poorly water-soluble components. In this study, we planned to mix the different oils, surfactants and co-surfactant to find out the self-emulsifying formulation with the highest oil and curcumin content, which is through detecting spontaneity, homogeneity and dispersibility of SEDS, Furthermore, it is necessary to investigate the effect of the product quality by using different curcumin loading on the quality of SEDS and the releasing contents of curcumin in the simulated gastrointestinal tract. The results show that Sorbitan monooleate(Span 80) has the best miscibility with algae oil, sunflower oil, olive oil and palm oil; the formulation with higher percentage of POE(20) Sorbitan Monooleate (Tween 80) have the best spontaneity, homogeneity and dispersibility; Tween 80 and Span 80 at a ratio of 1 : 1 as a surfactant formulation can have the product at the highest oil loading, and it can be mixed with palm oil to achieve the highest oil load 66% and the minimum particle size of 267.7 nm; In this formulation, the solubility of curcumin can be up to 6.37 mg/mL. In addition, adding different curcumin contents-3, 4.5 and 6 m/mL-to this SEDS formulation can be find that the increasing particle size and turbidity is correlated with the increase of curcumin loading. Also when we loaded 6 mg curcumin in the formulation, there will be the maximum particle size of 334.0 nm. In the simulated gastrointestinal release test, the release contents percentage of curcumin in simulated gastric fluid and intestinal fluid is about 69 and 80% respectively. In the follow-up research, we'll focus on how to reduce the release of curcumin in the stomach.
