簡易高效能液相層析法對市售薑黃素Curcumin產品之分析

摘要


本論文係建立一種對薑黃素 (curcumin) 進行分析的簡易高效能 液相層析分析法,進而應用於市售薑黃素保健食品之 curcumin 的含量 分析。本方法利用液相層析 C18 管柱(Alltech Alltima C18 column,4.6 ×150 mm,5μm),以乙腈/水(70/ 30,v/v)之混合溶液為移動相,流 速為 0.8 mL/min,於波長 420 nm 下進行偵測。 於基本分析方法上,檢量線範圍於 0.5〜100 μg/mL,其廻歸方程 式為 y = 145.76 x – 67.11,相關係數 r = 0.9998,顯示此方法定量性相 當良好。curcumin 偵測極限(LOD, S/N = 3)為 0.2 μg/mL,定量極限 (LOQ, S/N = 5)為 0.5 μg/mL。而同日內和異日間分析方法的精密度 和準確度,以相對標準偏差 (R.S.D.) 及相對誤差 (R.E.) 來表示,其 數值分別為小於 0.94% 和 4.61%(n = 6)。應用此建立之分析方法於 市售四種薑黃素保健產品的 curcumin 含量分析,其平均回收率為 99.3%,市售四種產品含量百分率分別為標示含量的 95.96 ± 0.98,15.49 ± 0.23,111.16 ± 1.26,21.35 ± 2.18。 本研究已建立一種分析 curcumin 之簡易 HPLC 方法,並成功地應 用在一些市售薑黃素保健食品的產品,分析其 curcumin 的含量。未來 本方法分析的結果,可以用來幫助社區藥局藥師建議民眾選擇合宜的市售薑黃素保健食品的一個參考依據。

The method includes reversed phase high performance liquid chromatography with ultraviolet detector was designed to determine curcumin in commercial products. Chromatography was performed by a C18 column (Alltech Alltima C18 column, 4.6 × 150 mm, 5 μm) and a mobile phase consisting of acetonitrile/water (70/30, v/v), flow rate was 0.8 mL/min. UV detection wavelength was at 420 nm. An isocratic HPLC method was applied to determine curcumin. Validation in selected conditions showed that the chosen method is sensitive, selective, precise and reproducible with linear response of detector for the determination of curcumin. The regression equation was y = 145.76 x – 67.11 between 0.5 ~ 100 μg/mL (r = 0.9998). The limit of detection (LOD, S/N = 3) for curcumin was 0.2 μg/mL. The limit of quantitation (LOQ, S/N = 5) was 0.5 μg/mL. The precision (as R.S.D.) and accuracy (as R.E.) of intraday and interday were less than 0.94% and 4.61%, respectively (n = 6). The average recovery of curcumin commercial products was 99.3%. The Mean ± S.D. percentage contents of curcumin in four commercial products A, B, C, D were 95.96 ± 0.98, 15.49 ± 0.23, 111.16 ± 1.26 and 21.35 ± 2.18 %, respectively. The method was also successfully applied to determine curcumin in some commercial curcumin products. In the future, the results of analysis can be used to help community pharmacists to recommend appropriate curcumin products in markets for the public.
