摘要


本研究的目的在於暸解鳳梨酵素對於心絲蟲成蟲體外之溶解能力,以了解未來運用於犬心絲蟲症臨床治療之可能性。實驗以PBS和血漿兩種溶劑及三種不同濃度的鳳梨酵素和對照組在37℃無菌環境下來觀察蟲體的溶解情形。結果顯示,以0.1 μg/mL Melarsomine dichlorhydrate殺死之蟲體於10 mg/mL鳳梨酵素的濃度下,公母蟲蟲體完全溶解的時間各為血漿♀27.0±1.1天,♂25.7±2.1天;PBS:♀26.3±1.9天,♂24.0±1.2天。在5 mg/mL的濃度下,完全溶解的時間各為血漿♀47.3±2.3天,♂43.7±2.3天;PBS:♀45.8±1.9天,♂42.2±1.9天(n=6);在1 mg/mL的濃度下則需60天以上。而對照組的蟲體完整性維持至6個月以上。由於鳳梨酵素主要辨認切割位置為Z-Arg-Arg + NHMec,因此推測蟲體含有類似之蛋白質結構。未來研究目標應朝向活體應用鳳梨酵素於心絲蟲症之研究。

The investigation was designed to examine the dissolving effect of bromelain on the canine adult heartworm body. Heartworms were killed by incubation with 0.1 μg/mL Melarsomine dichlorhydrate, incubated in respective designed mediums (Millipore-filtered) and solvent control at 37℃ for up to 2 months. Worm bodies incubated in 5 mg/mL (plasma: ♀ 47.3 ± 2.3; ♂ 43.7 ± 2.3; PBS: ♀ 45.8 ± 1.9; ♂ 42.2 ± 1.9 days) and 10 mg/mL (plasma: ♀27.0 ± 1.1; ♂ 25.7 ± 2.1; PBS: ♀ 26.3 ± 1.9; ♂ 24.0 ± 1.2 days) (n = 6) bromelain mediums were completely dissolved whereas those incubated in 1 mg/mL mediums exhibited body lysis only after 60 days. In tact worms structure was observed in solvent controls and lasted for 6 months onward. Dissolving velocity in both genders showed a similar pattern (P>0.05). Bromelain possesses more lysis potency in PBS mediums than those in plasma. It might be due to sera proteins which could reduce enzyme dissolving ability. Conclusively, bromelain is able to dissolve the heartworm in vitro. It suggests that heartworm contains Z-Arg-Arg + NHMec components. Future study should be toward on in vivo application of bromelain in canine dirofilariasis.
