含鞣花單寧之青草藥保肝活性生體外試驗

The In Vitro study of Ellagitannin-contained Herbs on the Hepatic Protection Activities

摘要


研究旨在篩選台灣民間常用含有韖花單寧(ellagitannin)之青草藥,並進行保肝活性之檢測,以期開發植物性保肝藥物。植物來源為屏東科技大學獸醫系藥園所種植者共157種之新鮮葉片,依ellagitannins之定性法進行薄層色層分析(TLC),取陽性者約48種之70%丙酮萃取的冷凍乾燥物,依本研究室建立之試管中大白鼠初代肝細胞培養模式,篩選出對四氯化碳所誘發的肝毒性有明顯抑制作用之青草藥,共20種。再以transaminase inhibition test確認其保肝活性。選出高抗肝毒性及低transaminase inhibition的植物有五種,即血桐,欖仁樹,老鸛草,水丁香及白飯樹。以上結果能做為生體內保肝活性檢定、進一步ellagitannin有效成分分離鑑定及其他生理活性檢測之參考依據。

The aim of this study was to evaluate hepatoprotective activities of the herbs which contain ellagitannins, and to develop herbal hepatoprotective drugs. One hundred and fifty seven kinds of herbs were collected from the herbal garden at Veterinary department in NPUST. The ellagitannin components were tested by using thin layer chromatography (TLC) method. There were 48 herb leaves showing positive ellagitannin coloration and the lyophilized powders of their fresh leaves extracted with 70% acetone were obtained. The extract samples were chosen for evaluation of antihepatotoxic activities by using an in-vitro rat primary cultural liver cell model. Twenty of the 48 herb samples showed antihepatotoxic activity against liver injury induced by carbon tetrachloride. In addition, the in-vitro antihepatotoxic activity was reconfirmed with a direct transaminase inhibition test. The results showed five herbs, namely, Macaranga tanarius (Linn.) Muell.-Arg., Terminalia catappa Linn., Geranium nepalense Sweet var. thunbergii (Sieb. & Zucco) Kudo, Ludwigia octovalvis (Jacq.) Raven., and Securinea virosa (Roxb.) Pax et Hoffm., had hepatoprotective activities, which were considered to have strong in-vitro antihepatotoxic activity with minimal transaminase inhibition.
