檢測家禽流行性感冒病毒非結構蛋白質抗體的間接型ELISA

Detecting Antibody to Avian Influenza Virus NS Protein by Indirect ELISA

摘要


由於施打疫苗的雞隻其血清中不含有NS1抗體,而感染野外株的雞隻會產生NS1抗體,故發展檢測NS1抗體之間接型ELISA(NS-ELISA)以區別雞隻體內的抗體的來源。從A/chicken/Taiwan/2838V/00(H6N1)病毒中抽取RNA,增幅NS1基因,在原核細胞表現NS1蛋白質,利用親和層分析法純化蛋白質,塗鍍於微平板做為抗原,發展檢測抗NS1抗體的ELISA。為防止NS1蛋白質中殘留之蛋白質干擾NS-ELISA的效果,以低齡及高齡雞隻的陰性血清以檢測訂定判定標準。結果顯示NS-ELISA可區分出陽性與陰性血清,且不受殘留之蛋白質干擾,其敏感性及特異性分別為94%及97%。故可用於區別雞隻體內的抗體是由施打疫苗或是感染野外毒產生的。

Due to no anti-NS1 antibody, an enzyme-linked immunosorbent assay (ELISA) using recombinant NS1 protein as an antigen (NS-ELISA) was developed to differentiate avian influenza virus (AIV)-infected from vaccinated chickens. The NS1-encoding gene of avian influenza A/chicken/Taiwan/2838V/00 (H6N1) virus was expressed in E. coli. The results showed that the sensitivity and specificity of this NS-ELISA were 94% and 97%, respectively. Therefore, this assay could be useful for serological diagnosis to distinguish poultry infected with influenza viruses from those immunized with inactivated vaccines.
