摘要

Pregabalin是作用在電壓閘控鈣離子通道上，對α2-δ受體具選擇性及高親和力的一種配體。其抑制性機轉可達成止痛、抗焦慮和抗癲癇的作用。許多研究已證實pregabalin在治療許多不同疾病上，如部分型癲癇、中樞及週邊神經性疼痛、泛焦慮症、不寧腿症和纖維肌痛症，具有卓著療效且安全性耐受度高。本文主要討論神經性疼痛和纖維肌痛症的治療，以及pregabalin相較gabapentin之優越性。

Pregabalin is a selective ligand with high affinity to theα_2-δsubunit of voltage-gated calcium channels. The inhibitory mechanism of pregabalin leads to the analgesic, anxiolytic and antiepileptic effects. Numerous controlled trials had demonstrated the advantageous efficacy, safety and tolerability of pregabalin in treatments for several distinct diseases e.g., partial epilepsy, central and peripheral neuropathic pain, generalized anxiety disorder, restless legs syndrome, and fibromyalgia. This article focuses on treatments of neuropathic pain and fibromyalgia, and the superiority of pregabalin comparing with gabapentin.
