針刺對腦出血模型大鼠急性期行為學及血腫周圍腦組織病理學的影響

Effect of Acupuncture on the Behavior of Guinea Pigs with Acute Cerebral Hemorrhage and Histopathology of Cerebral Hemotoma

Purpose: Research on the neurological deficit score, pathology of the cerebral hematoma, pattern of nerve cell damage of Guinea pigs with intracerebral hemorrhage and effects of acupuncture on NDS and nerve cell damage. Methods: 104 male SD Guinea pigs of 3 months old are divided randomly into control group of 8, psudo-surgery group of 32, hemorrhage group of 32, and hemorrhage with acupuncture of 32. The last three groups are randomly divided into 4 groups of 6h, 24h, 72, and 7d with 8 in each. Cerebral 3D Positioning instrument is used in control group and acupuncture group to inject 0.8U (0.1U/μl) collagen enzyme into right nuclei caudate to establish cerebral hemorrhage; acupuncture group receives acupuncture once a day on DaZhui, FengFu and ShuiGou. Behavioral and pathological methods are applied to observe the pathology of nerve function deficit and tissues around hematoma in Guinea pigs with acute cerebral hemorrhage. Same methods are used to observe the different patterns of nerve cells damage and effects of acupuncture on NDS and nerve cells damage. Result: (1) NDS: Group 6h shows deficit in nerve function immediately. The peak is reached after 48h and starts to drop at 72h. Group 7d is significantly higher than psudo-surgery group and control group. NDS values of 48h and 72h acupuncture groups are significantly lower than those of control group (P<0.05 and 0.01). This shows that acupuncture can improve nerve function in ICH Guinea pigs and has immediate awakening effect. (2) Pathomophology changes: In group 6h, it shows dissolved DIC with infiltration of neutrophils and formation of pus; dissolved red blood cells in hematoma with infiltration of neutrophils in group 24h; absorption of red blood cells, decreased infiltration of neutrophils, and visible filtration of lymphocytes and monocytes in group 72h; chronic inflammation and proliferation of fibro-connective tissues. (3) Effects of acupuncture on nerve cells damage: nerve cell damage was shown immediately in group 6h, reaches peak in 24h, last for 72h, and drops in 7d, but the extent is greater than those in control group. Numbers of damaged nerve cells in group 72h are greater than those in acupuncture group (p<0.01). Conclusion: Acupuncture can significantly improve deficit nerve function in ICH guinea pigs. There is infiltration of neutrophils in tissues around hematoma in acute ICH indicating severe damage of nerve cells. Acupuncture can decrease certain degree of nerve cell damage in ICH.
