Apple Polyphenol Inhibits Lipopolysaccharide-Induced Inflammation in an Animal Model

蘋果多酚在動物模式中抑制脂多醣體誘導之發炎作用

背景與目的:蘋果多酚在先前文獻中被證實具有抗氧化作用,為了進一步探究其抗發炎效用,脂多醣誘導動物肝臟發炎的模式在此被施行以進行研究。方法:蘋果多酚預先給予動物5天,五天後給予腹腔注射5 mg/kg脂多醣。6小時後犧牲動物,測定血漿中丙胺酸轉胺酶及天門冬胺酸轉胺酶(ALT及AST),以及檢查肝臟組織的變化。結果:本結果顯示在發炎模式動物中,蘋果多酚可降低血漿中ALT及AST,並同時改善肝組織中的抗氧化分子,包括增加catalase活性及glutathione(GSH)含量,並減少肝臟中的脂質過氧化現象。由組織切片也發現蘋果多酚可降低肝臟中LPS誘導之嗜中性球浸潤的現象。討論:目前結果暗示蘋果多酚在動物中具有抗發炎作用,並具潛力發展為抗發炎的物質。

Background and purpose: Previous studies demonstrate antioxidant effects of apple polyphenols (AP). In the present study, the potential anti-inflammatory effects of AP were examined in an animal model of lipopolysaccharide (LPS)-induced hepatic inflammation.Methods: Rats were pretreated with AP for five days before intraperitonial(i.p.) administration of 5 mg/kg LPS. After 6 h, the animals were sacrificed, and plasma alanine (ALT) levels, aspartate aminotransferase (AST) levels, glutathione (GSH) levels, catalase activity, and lipid peroxidation were assessed in liver tissues.Results: Pretreatments with AP decreased hepatic LPS-induced ALT and AST levels. Moreover AP pretreatment preserved antioxidant catalase activity and glutathione (GSH) levels and decreased lipid peroxidation in the livers of LPS-treated rats. Histopathological evaluations of rat livers revealed that AP reduced the incidence of LPS-induced liver lesions and neutrophil infiltration.Discussion: The present data indicate that AP has anti-inflammatory potential in vivo and could be developed as an anti-inflammatory agent.
