台灣地區漢族之STR基因分析研究-Investigator 24plex QS Kit之應用

摘要


本研究使用Investigator 24plex QS Kit試劑組分析303位台灣地區隨機漢人在21個體染色體、1個Y染色體STR基因之分布情形,包含CSF1PO、D2S1338、D3S1358、D5S818、D7S820、D8S1179、D13S317、D16S539、D18S511、D19S433、D21S11、FGA、TH01、TPOX、vWA、D1S1656、D2S441、D22S1045、D10S1248、D12S391、SE33與DYS391。此分析系統亦同時分析Amelogenin與一對品管基因。由於目前常用之CODIS 15個STR基因系統在二人組親子排除率無法達到親子鑑定規範之要求,在人別鑑定時亦有隨機相符機率不足要求與混合型判讀限制之現象,因此增加篩檢STR基因成為鑑識實驗室提高鑑別率之重要策略。本系統亦同時複製一對品管基因,可評估檢體品質。在一致性比對中,經與GlobalFiler^(TM) Express PCR Amplification Kit分析之結果比較,在相同STR基因之基因型均完全一致。依此系統之21個體染色體STR基因數據計算此族群綜合隨機相符機率、最常見型相符機率、二人組親子排除率與三人組親子排除率,其累積值分別是2.58E-25、2.96E-19、99.9997%與99.99999990%,此排除率數據可符合現行親緣鑑定技術規範之要求。

This study screened 303 random individuals in Taiwan Han population using Investigator 24plex QS Kit including 21 STR loci in autosome, CSF1PO, D2S1338, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S511, D19S433, D21S11, FGA, TH01, TPOX, vWA, D1S1656, D2S441, D22S1045, D10S1248, D12S391, SE33 and 1 Y-STR locus DYS391 for their distributions. This system also screened Amelogenin gene and a pair of quality sensors. Since the power of exclusion obtained using 15 STR loci was not met the requirements of the guideline of relationship test in duo case and resolved the mixture problem in individual identification, more polymorphic markers were needed to increase its discrimination power. The quality sensors in this system can evaluate the quality of samples. In the concordance study, all samples have same genotypes comparing with those genotyped using GlobalFiler^(TM) Express PCR Amplification Kit. The random matching probability, matching probability of the most popular genotype, power of exclusion in duo and trio case using this system were 2.58E-25, 2.96E-19, 99.9997% and 99.99999990% respectively. This power of exclusion is able to meet of the requirements of current guideline in relationship testing.
