鹼催化對Folin-Ciocalteu試劑檢測總多酚含量的影響

摘要


Folin-Ciocalteu比色法是最普遍應用於檢測抗氧化總多酚含量的方法,定量上常以沒食子酸當量作為總多酚相對含量的表示。本研究討論Folin-Ciocalteu比色法中碳酸鈉添加對檢測總多酚的影響。結果表明,沒食子酸在鹼性環境下具有化學的不穩定性,當20%碳酸鈉添加的步驟在Folin-Ciocalteu試劑之前,沒食子酸因鹼而形成不可逆的化合物,直接影響到Folin-Ciocalteu試劑的還原反應效率,進而對總多酚定量檢測造成顯著的干擾。

Gallic acid is often used as the quantitative standards in Folin-Ciocalteu colorimetric method for determination of total polyphenols. The evaluation of total polyphenol content is equivalent to the relative content of gallic acid. The effects of sodium carbonate on total polyphenol analysis with Folin-Ciocalteu reagent were studied.The results showed that gallic acid is unstable in alkaline solution. When 20% sodium carbonate was added before the addition of Folin-Ciocalteu reagent in the assay, an irreversible compound had been produced. With the alteration of reaction conditions, the electric transferring capacity of Folin-Ciocalteu reagent was decreased during its reduction. Therefore, the quantitative analyses of total polyphenols and calibration of using gallic acid as standards were also shown to be affected significantly.
